辣椒辣素和苯酚對小鼠變應(yīng)性接觸性皮炎影響的研究

【摘要】:目的: 觀察辣椒辣素和苯酚對小鼠變應(yīng)性接觸性皮炎的影響,探討神經(jīng)系統(tǒng)與變應(yīng)性接觸性皮炎的關(guān)系和可能機理。 方法: 選用雄性清潔級BALB/c小鼠48只,6-8周齡,將小鼠隨機分為12組,以2,4-二硝基氯苯作為致敏劑于小鼠腹部致敏、右耳激發(fā)建立小鼠ACD模型。選用小鼠雙側(cè)耳厚差、耳重量差、組織切片顯微鏡下耳雙面距離差、組織切片中浸潤細胞的種類及數(shù)量、雙側(cè)耳引流淋巴結(jié)中淋巴細胞的增值活性等指標(biāo)對小鼠ACD進行評價。實驗①分別用特異性肽能神經(jīng)毒性物質(zhì)辣椒辣素和一般神經(jīng)毒性化合物苯酚對小鼠行致敏前或激發(fā)前預(yù)處理,觀察辣椒辣素和苯酚對ACD小鼠的傳入支(致敏階段)或傳出支(激發(fā)階段)的影響; 實驗②在致敏前2天使用辣椒辣素,并于致敏后3天、6天、9天、12天、15天再激發(fā),觀察辣椒辣素對小鼠ACD的影響與時間的關(guān)系。 結(jié)果: 1.小鼠ACD模型造模成功。模型組及各對照組小鼠雙側(cè)耳厚差、耳重量差、組織切片顯微鏡下耳雙面距離差、單一核細胞數(shù)及多形核數(shù)目差及雙側(cè)耳后淋巴結(jié)中淋巴細胞的增值活性相比均有統(tǒng)計學(xué)差異(p0.01); 2.致敏前使用辣椒辣素或苯酚使局部皮膚神經(jīng)纖維失活,或者系統(tǒng)性地使肽能神經(jīng)失活后,雙側(cè)耳的各指標(biāo)無統(tǒng)計學(xué)差異(p0.05),激發(fā)前用辣椒辣素使局部皮膚神經(jīng)纖維失活,雙側(cè)耳的各指標(biāo)無統(tǒng)計學(xué)差異(p0.05); 3.致敏前2天使用辣椒辣素,并于致敏后3天、6天、9天、12天、15天再激發(fā),雙側(cè)耳的各指標(biāo)的差異無統(tǒng)計學(xué)意義(p0.05)。 結(jié)論: 1.辣椒辣素能阻止變應(yīng)性接觸性皮炎的形成,其機理是通過阻斷小鼠變應(yīng)性接觸性皮炎的傳入支(致敏階段)和傳出支(激發(fā)階段)。 2.苯酚能阻斷小鼠變應(yīng)性接觸性皮炎的傳入支(致敏階段)。 3.神經(jīng)系統(tǒng)在小鼠變應(yīng)性接觸性皮炎的發(fā)生和發(fā)展中起著重要的作用。
[Abstract]:Objective: To observe the effects of capsaicin and phenol on allergic contact dermatitis in mice and to explore the relationship between nervous system and allergic contact dermatitis and its possible mechanism. Methods: Forty-eight male clean grade BALB/c mice aged 6-8 weeks were randomly divided into 12 groups. The mice were sensitized by 2-di-4-dinitrochlorobenzene in abdomen, and ACD model was established by right ear stimulation. The ACD of mice was evaluated by using the indexes of poor ear thickness, poor ear weight, the distance between the two sides of the ear under the microscope, the type and number of infiltrating cells in the tissue sections, and the proliferative activity of lymphocytes in the lymph nodes of the bilateral lateral ear drainage. In experiment 1, mice were pretreated with capsaicin, a specific peptidergic neurotoxic substance, and phenol, a general neurotoxic compound, before or before stimulation. The effects of capsaicin and phenol on the afferent branch (sensitizing stage) and efferent branch (excitation stage) of ACD mice were observed. In experiment 2, capsaicin was used 2 days before sensitization, and then stimulated 3 days, 6 days, 9 days, 12 days and 15 days after sensitization. The effect of capsaicin on ACD in mice was observed. Results: 1. Mouse ACD model was successfully established. In the model group and the control group, there were significant differences in the thickness of the ears, the weight of the ears, the distance between the two sides, the number of mononuclear cells and the number of polymorphic nuclei, and the proliferative activity of lymphocytes in the lymph nodes of the posterior lymph nodes of the two lateral ears. 2. After using capsaicin or phenol to inactivate local skin nerve fibers or systematically inactivating peptidergic nerves before sensitization, there was no significant difference in the parameters of the two lateral ears (p 0.05), and capsaicin was used to inactivate local skin nerve fibers before sensitization. There was no statistical difference in each index of the two lateral ears (p0.05). 3. Capsaicin was used 2 days before sensitizing, and was stimulated again 3 days, 6 days, 9 days, 12 days and 15 days after sensitization. There was no significant difference in the indexes of two lateral ears (P 0.05). Conclusion: 1. Capsaicin can prevent the formation of allergic contact dermatitis by blocking the afferent branch (sensitizing stage) and efferent branch (excitation stage) of allergic contact dermatitis in mice. 2. Phenol can block the afferent branches of allergic contact dermatitis in mice. 3. Nervous system plays an important role in the occurrence and development of allergic contact dermatitis in mice.
