【摘要】：銀屑病是由環(huán)境因素激發(fā)的、遺傳因素決定的慢性炎癥皮膚病。而局部微環(huán)境的缺氧和表皮角質(zhì)形成細(xì)胞的增殖是此疾病最重要的特征。我們的前期實(shí)驗(yàn)已經(jīng)證明CCL20和HIF-1α在銀屑病患者病損區(qū)活檢組織中的表達(dá)較正常人明顯增強(qiáng)。但是目前對(duì)CCL20在缺氧微環(huán)境中上調(diào)的機(jī)制還不甚明白。 通過(guò)ELISA技術(shù)檢測(cè)17例銀屑病患者與6例正常人血清中CCL20水平后發(fā)現(xiàn): CCL20在銀屑病患者血清中的表達(dá)水平(26.76±19.34pg/ml)顯著高于正常對(duì)照組(8.69±3.19 pg/ml),增高約3倍,有統(tǒng)計(jì)學(xué)意義,P0.05。 本試驗(yàn)選用Hacat細(xì)胞作為正常角質(zhì)形成細(xì)胞(KC)的模型,利用CO2培養(yǎng)箱(94%N2, 5%CO2, 1%O2)模擬銀屑病患者皮損中局部缺氧微環(huán)境。通過(guò)RT-PCR技術(shù)檢測(cè)發(fā)現(xiàn)表皮角質(zhì)形成細(xì)胞缺氧6-12小時(shí)后,CCL20和HIF-1α表達(dá)明顯增高,隨后漸回到基線水平。此外我們還發(fā)現(xiàn)CCL20和HIF-1α的表達(dá)呈正相關(guān)(r = 0.855,p0.05)。因?yàn)閷⑷毖鹾蟮谋砥そ琴|(zhì)形成細(xì)胞轉(zhuǎn)染HIF-1αsiRNA下調(diào)HIF-1α表達(dá)后,CCL20在mRNA及蛋白水平的表達(dá)均回到基礎(chǔ)水平。 綜上所述,CCL20可能在銀屑病發(fā)病的免疫機(jī)制中發(fā)揮著重要的作用,CCL20的上調(diào)可能與HIF-1α有關(guān),并且兩者在銀屑病發(fā)病中可能起協(xié)同作用。
[Abstract]:Psoriasis is a chronic inflammatory dermatosis caused by environmental factors and genetic factors. Hypoxia in local microenvironment and proliferation of epidermal keratinocytes are the most important characteristics of the disease. Our previous experiments have demonstrated that the expression of CCL20 and HIF-1 偽 in biopsy tissues of psoriatic lesions was significantly higher than that in normal controls. However, the mechanism of CCL20 upregulation in hypoxia microenvironment is not well understood. The expression of CCL20 in serum of 17 patients with psoriasis and 6 normal controls was detected by ELISA technique. It was found that the expression level of CCL20 in serum of psoriasis patients was significantly higher than that of normal controls (P = 26.76 鹵19.34pg / ml) and 8.69 鹵3.19pg / ml respectively, which was about 3 times higher than that of normal controls (P 0.05). In this experiment, Hacat cells were used as the model of normal keratinocytes (KC), and N2,5CO2,1O2 were used in CO2 incubator to simulate the local anoxic microenvironment in the skin lesions of psoriasis patients. The expression of CCL20 and HIF-1 偽 in epidermal keratinocytes was significantly increased after hypoxia for 6-12 hours, and then returned to baseline level. In addition, we also found a positive correlation between the expression of CCL20 and HIF-1 偽 (r = 0.855P 0.05). The expression of CCL20 in mRNA and protein levels of HIF-1 偽 siRNA was reduced to the basic level after transfection of hypoxia-induced epidermal keratinocytes (EKCs) to down-regulate the expression of HIF-1 偽. In conclusion, CCL20 may play an important role in the immune mechanism of psoriasis. The up-regulation of CCL20 may be related to HIF-1 偽, and both may play a synergistic role in the pathogenesis of psoriasis.
