Toll樣9受體單核苷酸多態(tài)性與尖銳濕疣感染相關性的初步研究

【摘要】: 尖銳濕疣是由人乳頭瘤病毒感染引起的一種性感染疾病,發(fā)病率高,已居性感染疾病的第二位。該病傳染性強,復發(fā)率高,對人類身心危害較大。近年來研究發(fā)現,免疫失衡在尖銳濕疣的發(fā)生、發(fā)展及轉歸等方面發(fā)揮重要作用。Toll樣受體是一類天然識別受體,在天然免疫和適應性免疫中發(fā)揮著重要的橋梁作用。Toll樣受體9(Toll like receptor 9,SNP)是其中的一種,主要識別未甲基化的CpG基序,從而在抗病毒免疫和自身免疫方面發(fā)揮重要的作用。本課題著重研究了尖銳濕疣患者外周血TLR9的單核苷酸多態(tài)性(Singlenucleotide polymorphism,SNP),分析Toll樣受體9的SNP與尖銳濕疣發(fā)病的相關性。 研究目的 探討Toll樣受體9的單核苷酸多態(tài)性與尖銳濕疣發(fā)病的相關性。 研究方法 抽取63例確診的尖銳濕疣患者和23例健康對照組的外周靜脈血,采用TLR9基因直接測序的方法分析TLR9受體的單核苷酸多態(tài)性。 結果 1.TLR9基因自翻譯起始點第+1174、1635、1269、1724位均存在單核苷酸多態(tài)性位點,分別稱為SNP1、SNP2、SNP3和SNP4,其中SNP3、SNP4為新發(fā)現位點,SNP1、SNP2為NCBI數據庫中公布的位點,登錄號分別為:rs352139、rs352140。SNP1、SNP2在人群中均可表現為三種基因型:AA純合、AG雜合和GG純合,這兩個位點的三種基因型在尖銳濕疣患者和健康對照組間比較差異均無統(tǒng)計學意義(P0.05)。SNP3、SNP4均只在1例尖銳濕疣患者中發(fā)現,均表現為AG雜合,其余個體該兩位點均為GG純合。 2.尖銳濕疣患者與健康對照組的SNP1位點的等位基因A的頻率分別為0.690、0.609,等位基因G的頻率0.309、0.391,兩組間比較差異均無統(tǒng)計學意義(P0.05)。SNP2位點的等位基因A的頻率分別為0.302、0.369,等位基因G的頻率0.698、0.630,兩組間比較差異無統(tǒng)計學意義(P0.05)。 3.SNP1、SNP2兩個多態(tài)性位點存在4種單倍體型,分別為AA、AG、GA、GG。每種單倍體型在尖銳濕疣患者組和健康對照組間比較差異均無統(tǒng)計學意義(P0.05)。 4.尖銳濕疣患者初發(fā)組與復發(fā)組的CD3+分別為:63.24%±10.09%、67.11%±9.28%,兩組間比較差異均無統(tǒng)計學意義(P0.05);CD4+分別為:32.13%±8.65%、30.89%±5.81%,兩組間比較差異均無統(tǒng)計學意義(P0.05);CD8+分別為:29.04%±9.29%、33.00%±8.06%,兩組間比較差異均無統(tǒng)計學意義(P0.05);Th/Ts分別為:1.25±0.53、1.00±0.33,兩組間差異具有統(tǒng)計學意義(P0.05)。 5.SNP1位點AA、AG、GG三種基因型的Th/Ts分別為:1.08±0.48、0.92±0.35、1.09±0.47,三種基因型間比較差異無統(tǒng)計學意義(P0.05);SNP2位點AA、AG、GG三種基因型的Th/Ts分別為:1.08±0.42、0.92±0.37、1.08±0.48,三種基因型間比較差異無統(tǒng)計學意義(P0.05)。 結論 1.TLR9基因在廣東漢族人群中存在4個單核苷酸多態(tài)性位點,分別為SNP1、SNP2、SNP3、SNP4。 2.SNP1、SNP2兩位點均與尖銳濕疣的發(fā)病易感性可能無相關性。 3.初發(fā)與復發(fā)的尖銳濕疣患者間存在T淋巴細胞亞群免疫失衡。 4.SNP1、SNP2兩位點與尖銳濕疣患者的T淋巴細胞亞群間可能無相關性。
[Abstract]:Toll like receptor 9 ( SNP ) is a kind of natural recognition receptor , which plays an important role in the pathogenesis , development and prognosis of condyloma . Toll like receptor 9 ( toll like receptor 9 , SNP ) plays an important role in innate immune and adaptive immunity . Toll like receptor 9 ( SNP ) is a kind of natural recognition receptor . Toll like receptor 9 ( SNP ) plays an important role in antiviral immunity and autoimmune diseases .
Purpose of study
To investigate the correlation between single nucleotide polymorphism of Toll - like receptor 9 and the pathogenesis of Condyloma Acuminatum .
Research Methods
The single nucleotide polymorphism of TLR9 receptor was analyzed by direct sequencing of TLR9 gene in 63 patients with acute condyloma and 23 healthy controls .
Results
1 . There were single nucleotide polymorphisms at the beginning of translation initiation point + 1174 , 1635 , 1269 and 1724 of TLR9 gene . SNP3 and SNP4 were identified as SNP1 , SNP2 , SNP3 and SNP4 , respectively .
2 . The frequencies of allele A were 0.690 , 0.609 , 0.309 and 0.391 , respectively , and there was no significant difference between the two groups ( P0.05 ) . The frequencies of allele A in SNP2 locus were 0.302 , 0.369 and 0.698 , 0.6630 , respectively . There was no significant difference between the two groups ( P0.05 ) .
3 . There were four haploid types of SNP1 and SNP2 polymorphic loci , AA , AG , GA and GG respectively . There was no significant difference between the two groups ( P0.05 ) .
4 . CD3 + was 63.24 % 鹵 10.09 % , 67.11 % 鹵 9.28 % in the primary and recurrent group , and there was no significant difference between the two groups ( P0.05 ) .
CD4 + was 32.13 % 鹵 8.65 % , 30.89 % 鹵 5.81 % , and there was no significant difference between the two groups ( P0.05 ) .
CD8 + was 29.04 % 鹵 9.29 % , 33.00 % 鹵 8.06 % , and there was no significant difference between the two groups ( P0.05 ) .
Th / Ts were 1.25 鹵 0.53 , 1.00 鹵 0.33 respectively . There was significant difference between the two groups ( P0.05 ) .
5 . Th / Ts of three genotypes of SNP1 locus AA , AG and GG were 1.08 鹵 0.48 , 0.92 鹵 0.35 , 1.09 鹵 0.47 respectively . There was no significant difference between the three genotypes ( P0.05 ) .
The Th / Ts of three genotypes of SNP2 loci AA , AG and GG were 1.08 鹵 0.42 , 0.92 鹵 0.37 , 1.08 鹵 0.48 respectively . There was no significant difference between the three genotypes ( P0.05 ) .
Conclusion
1 . The TLR9 gene had four single nucleotide polymorphism sites in Guangdong Han population , SNP1 , SNP2 , SNP3 , SNP4 , respectively .
2 . Both SNP1 and SNP2 had no correlation with the susceptibility to condyloma .
3 . There was an imbalance of T - lymphocyte subsets in patients with primary and recurrent acuminate condyloma .
4 . There may be no correlation between SNP1 , SNP2 and T - lymphocyte subsets in patients with Condyloma Acuminatum .
