強(qiáng)脈沖光治療紫外線所致皮膚光老化作用的機(jī)制研究

【摘要】: 探討強(qiáng)脈沖光治療紫外線所致皮膚光老化的可能作用機(jī)制,為臨床應(yīng)用提供理論依據(jù)。 30只大鼠隨機(jī)分為三組:正常對(duì)照組,UV照射組,IPL治療組。正常組不施加任何處理因素,僅同步喂養(yǎng);紫外線照射組僅照射紫外線;IPL治療組為紫外線照射+IPL照射。采用HE染色觀察各組皮膚組織病理改變,檢測(cè)各組皮膚組織中MDA含量,SOD、GSH-pX、CAT活性,western blot檢測(cè)皮膚組織中MMP-1、TGF--β1以及bFGF表達(dá)變化。 (1)正常對(duì)照組大鼠表皮組織結(jié)構(gòu)完整,細(xì)胞分層清晰,真皮內(nèi)彈性纖維和膠原纖維形態(tài)及走向正常;紫外線照射后,大鼠的表皮組織變薄,結(jié)構(gòu)不甚完整,分層不清,表皮與真皮交界呈直線狀,纖維組織發(fā)生變性、破壞,可見(jiàn)真皮組織排列紊亂,分布不均或稀疏,有較多的炎性細(xì)胞浸潤(rùn);IPL治療組大鼠皮膚組織受損程度較輕,表皮組織結(jié)構(gòu)比較完整,細(xì)胞分層比較清晰,皮膚厚薄不甚均一,真皮內(nèi)彈性纖維和膠原纖維形態(tài)及走向基本正常。 (2)UV照射組大鼠皮膚組織中MDA含量較正常皮膚組織顯著增加,而皮膚組織中SOD、GSH-pX、CAT含量較正常皮膚組織顯著降低,P0.05。IPL治療后,大鼠皮膚組織中MDA含量較UV照射組大鼠顯著降低,而皮膚組織中SOD、GSH-pX、CAT含量較UV照射組大鼠顯著增高,P0.05。 (3)UV照射組大鼠皮膚組織中MMP-1蛋白表達(dá)較正常皮膚組織顯著增加,而皮膚組織中TGF-β1、bFGF含量較正常皮膚組織顯著降低,P0.05。IPL治療后,大鼠皮膚組織中MMP-1蛋白表達(dá)較UV照射組大鼠顯著降低,而皮膚組織中TGF-β1、bFGF蛋白表達(dá)較UV照射組大鼠顯著增高,P0.05。 (1)長(zhǎng)期紫外線照射導(dǎo)致大鼠發(fā)生光老化。 (2)長(zhǎng)期紫外線照射可致皮膚組織中產(chǎn)生脂質(zhì)過(guò)氧化而致氧化應(yīng)激,使MDA含量升高,SOD、GSH-px、CAT活性降低,可致皮膚組織中MMP-1蛋白含量增高。 (3)IPL可治療長(zhǎng)期紫外線輻射所致的光老化,改善皮膚的外觀,降低皮膚組織中MDA含量,提高SOD、GSH-px、CAT活性。同時(shí)可使皮膚組織中MMP-1蛋白含量降低,增加TGF-β和bFGF蛋白含量。 (4)調(diào)節(jié)機(jī)體氧化損傷與抗氧化損傷動(dòng)態(tài)的平衡,通過(guò)誘導(dǎo)有損傷修復(fù)作用的細(xì)胞因子TGF-β和bFGF表達(dá),抑制參與皮膚組織膠原降解的MMP-1表達(dá),可能是IPL治療紫外線所致皮膚光老化的機(jī)制之一。
[Abstract]:To explore the possible mechanism of intense pulse light in treating photoaging of skin induced by ultraviolet ray, and to provide theoretical basis for clinical application.Thirty rats were randomly divided into three groups: normal control group treated with IPL.No treatment factors were applied in the normal group, and only simultaneous feeding was performed in the ultraviolet radiation group, and IPL irradiation was only used in the ultraviolet irradiation group.The pathological changes of skin tissue were observed by HE staining. The content of MDA and the activity of GSH-pXG cat in skin tissue were detected. The expression of MMP-1TGF- 尾 1 and bFGF in skin tissue were detected by western blot.1) in normal control group, the structure of epidermal tissue was intact, the layer of cells was clear, the shape and direction of elastic fiber and collagen fiber in dermis were normal, the epidermal tissue of rats was thinned, the structure was not complete, and the layer was not clear after ultraviolet irradiation.The junction of epidermis and dermis was linear, the fibrous tissue was degenerated and destroyed, the dermis tissue was arranged in disorder, distributed unevenly or sparsely, and there were more inflammatory cells infiltrating into the skin tissue of IPL treatment group, and the damage degree of skin tissue in IPL treatment group was less than that in IPL group.The structure of epidermis was relatively complete, the layer of cells was clear, the thickness of skin was not uniform, and the morphology and direction of elastic fibers and collagen fibers in dermis were normal.The content of MDA in skin tissue was significantly higher than that in normal skin tissue but the content of MDA in skin tissue was significantly lower than that in normal skin tissue after treatment with P0.05. IPL the content of MDA in skin tissue was significantly lower than that in UV irradiated group.The content of GSH-pXX cat in skin tissue was significantly higher than that in UV irradiation group (P 0.05).The expression of MMP-1 protein in the skin tissue was significantly increased than that in the normal skin tissue while the content of TGF- 尾 1 bFGF in the skin tissue was significantly lower than that in the normal skin tissue after treatment with P0.05. IPL the expression of MMP-1 protein in the skin tissue of the rats was significantly lower than that in the UV irradiated group.The expression of TGF- 尾 1 bFGF protein in skin tissue was significantly higher than that in UV irradiation group (P 0.05).Long-term ultraviolet radiation induced photoaging in rats.(2) Long-term ultraviolet irradiation could induce lipid peroxidation and oxidative stress in skin tissue, increase the content of MDA, decrease the activity of GSH-pxX cat, and increase the content of MMP-1 protein in skin tissue.It can treat the photoaging caused by ultraviolet radiation for a long time, improve the appearance of skin, decrease the content of MDA in skin tissue and increase the activity of GSH-pxX cat.At the same time, the content of MMP-1 protein in skin tissue was decreased, and the content of TGF- 尾 and bFGF protein was increased.(4) regulating the dynamic balance between oxidative and antioxidant injury, inhibiting the expression of MMP-1 involved in collagen degradation by inducing the expression of TGF- 尾 and bFGF.It may be one of the mechanisms of IPL in the treatment of ultraviolet-induced skin photoaging.
