【摘要】：目的研究阿維A調(diào)控銀屑病患者外周血樹突狀細胞功能,探討阿維A治療銀屑病作用機制。方法抽取銀屑病患者外周血,分離單個核細胞,體外誘導培養(yǎng)樹交狀細胞(Dendritic cells,DCs)。不同濃度阿維A(Inmol/L和10nmol/L)作用DCs 24h,對照組為只加培養(yǎng)基,流式細胞術(shù)(Flow cytometry,FCM)檢測DCs中CD80,CD83及CD86表達水平,酶聯(lián)免疫吸附試驗(ELISA)方法檢測細胞培養(yǎng)上清液中IL-12表達水平。結(jié)果體外成功培養(yǎng)出DCs,流式細胞術(shù)檢測到1,10nmol/L阿維A作用DCs24h后,CD80,CD83及CD86表達率分別為(69.74±3.83)%、(61.12±3.03)%、(70.18±3.32)%和(60.19±3.97)%、(50.78±4.81)%、(48.96±3.18)%,顯著低于對照組DCs上CD80,CD83及CD86表達率(75.15±2.76)%、(72.43±1.94)%、(77.81±2.46)%(P0.01)。1,10nmol/L阿維A作用DCs 24h后,DCs分泌到細胞培養(yǎng)液中的IL-12表達水平分別為(260.36±15.26)(226.98±17.24)顯著低于對照組(282.80±23.26)(P0.01)。結(jié)論阿維A可以降低銀屑病患者外周血中DCs上CD80.CD83及CD86表達率,抑制DCs活化,且降低IL-12因子的分泌量,能夠調(diào)控患者機體的免疫功能,從而對銀屑病起到治療作用。
[Abstract]:Objective to study the regulation of Avera on the function of peripheral blood dendritic cells in patients with psoriasis and to explore the mechanism of Avera in the treatment of psoriasis.Methods Mononuclear cells were isolated from peripheral blood of patients with psoriasis, and dendritic cells were cultured in vitro.DCs was treated with Avi A(Inmol/L and 10nmol / L for 24 h. Flow cytometry was used to detect the expression of CD80 CD83 and CD86 in DCs. Elisa was used to detect the expression of IL-12 in the supernatant of cell culture.
