【摘要】：目的:測定皮膚鱗狀細胞癌(CSCC)組織中人乳頭瘤病毒(HPV)、miR-21及miR-31的表達量,分析HPV感染與miRNA之間的相關(guān)性,并探討miR-31可能的致癌機制。方法:采用HPV DNA檢測試劑盒和巢式聚合酶鏈?zhǔn)椒磻?yīng)(PCR)檢測CSCC組和正常對照組中HPV表達情況,實時熒光定量PCR分別測定HPV陽性和陰性CSCC組織中miR-21及miR-31的表達量,免疫組化檢測CSCC組織中Ras GTP酶激活蛋白(RASA)1表達水平。結(jié)果:CSCC組織中HPV檢出率為18.29%(15/82)。miR-21和miR-31在CSCC組織中相對表達量分別為0.2750±0.2280和0.0130±0.0220,均高于正常對照組(0.0110±0.0190、0.0002±0.0002),差異均有統(tǒng)計學(xué)意義(P均0.05);RASA1蛋白在CSCC組織中表達明顯減少(P0.05)。在HPV陰性組織中RASA1蛋白表達與miR-31水平呈負相關(guān)(r=-0.588,P0.05)。結(jié)論:HPV可能在CSCC發(fā)展早期起一定作用;miR-31可能是HPV相關(guān)性miRNA;高表達的miR-31可能通過下調(diào)RASA1促進細胞增殖,參與CSCC的發(fā)生及發(fā)展。
[Abstract]:Objective: to detect the expression of human papillomavirus (HPVV) miR-21 and human papillomavirus (miR-31) in skin squamous cell carcinoma (CSCC) tissues, and to analyze the correlation between HPV infection and miRNA. Methods: HPV DNA assay kit and nested polymerase chain reaction (PCR) were used to detect the expression of HPV in CSCC group and normal control group. Real-time fluorescence quantitative PCR was used to detect the expression of miR-21 and miR-31 in HPV positive and negative CSCC tissues. Immunohistochemistry was used to detect the expression of Ras GTP enzyme activator protein (RASA) 1 in CSCC. Results the relative expression levels of 18.29%(15/82).miR-21 and miR-31 in CSCC were 0.2750 鹵0.2280 and 0.0130 鹵0.0220, respectively, which were significantly higher than those in normal control group (0.0110 鹵0.0190,0.0002 鹵0.0002). In HPV negative tissues, the expression of RASA1 protein was negatively correlated with the level of miR-31. Conclusion CSCC may play a role in HPV related miRNAs in early stage of CSCC development, and high expression of miR-31 may play a role in the development of CSCC. Can promote cell proliferation by down-regulating RASA1, Participate in the occurrence and development of CSCC.
