【摘要】：目的:測(cè)定皮膚鱗狀細(xì)胞癌(CSCC)組織中人乳頭瘤病毒(HPV)、miR-21及miR-31的表達(dá)量,分析HPV感染與miRNA之間的相關(guān)性,并探討miR-31可能的致癌機(jī)制。方法:采用HPV DNA檢測(cè)試劑盒和巢式聚合酶鏈?zhǔn)椒磻?yīng)(PCR)檢測(cè)CSCC組和正常對(duì)照組中HPV表達(dá)情況,實(shí)時(shí)熒光定量PCR分別測(cè)定HPV陽(yáng)性和陰性CSCC組織中miR-21及miR-31的表達(dá)量,免疫組化檢測(cè)CSCC組織中Ras GTP酶激活蛋白(RASA)1表達(dá)水平。結(jié)果:CSCC組織中HPV檢出率為18.29%(15/82)。miR-21和miR-31在CSCC組織中相對(duì)表達(dá)量分別為0.2750±0.2280和0.0130±0.0220,均高于正常對(duì)照組(0.0110±0.0190、0.0002±0.0002),差異均有統(tǒng)計(jì)學(xué)意義(P均0.05);RASA1蛋白在CSCC組織中表達(dá)明顯減少(P0.05)。在HPV陰性組織中RASA1蛋白表達(dá)與miR-31水平呈負(fù)相關(guān)(r=-0.588,P0.05)。結(jié)論:HPV可能在CSCC發(fā)展早期起一定作用;miR-31可能是HPV相關(guān)性miRNA;高表達(dá)的miR-31可能通過(guò)下調(diào)RASA1促進(jìn)細(xì)胞增殖,參與CSCC的發(fā)生及發(fā)展。
[Abstract]:Objective: to detect the expression of human papillomavirus (HPVV) miR-21 and human papillomavirus (miR-31) in skin squamous cell carcinoma (CSCC) tissues, and to analyze the correlation between HPV infection and miRNA. Methods: HPV DNA assay kit and nested polymerase chain reaction (PCR) were used to detect the expression of HPV in CSCC group and normal control group. Real-time fluorescence quantitative PCR was used to detect the expression of miR-21 and miR-31 in HPV positive and negative CSCC tissues. Immunohistochemistry was used to detect the expression of Ras GTP enzyme activator protein (RASA) 1 in CSCC. Results the relative expression levels of 18.29%(15/82).miR-21 and miR-31 in CSCC were 0.2750 鹵0.2280 and 0.0130 鹵0.0220, respectively, which were significantly higher than those in normal control group (0.0110 鹵0.0190,0.0002 鹵0.0002). In HPV negative tissues, the expression of RASA1 protein was negatively correlated with the level of miR-31. Conclusion CSCC may play a role in HPV related miRNAs in early stage of CSCC development, and high expression of miR-31 may play a role in the development of CSCC. Can promote cell proliferation by down-regulating RASA1, Participate in the occurrence and development of CSCC.
