【摘要】：目的：考察納米雄黃對(duì)皮膚鱗癌A431細(xì)胞凋亡的作用，探討納米雄黃誘導(dǎo)腫瘤細(xì)胞凋亡和抑制增殖的機(jī)理。方法：本實(shí)驗(yàn)主要包括納米雄黃通過(guò)機(jī)械研制法制備，A431細(xì)胞的形態(tài)學(xué)觀察，運(yùn)用免疫組化法、RT-PCR技術(shù)探討納米雄黃誘導(dǎo)人皮膚鱗癌A431細(xì)胞凋亡和抑制增殖的作用機(jī)理。結(jié)果：納米雄黃可以明顯誘導(dǎo)人皮膚鱗癌A431細(xì)胞的凋亡，且與順鉑聯(lián)合應(yīng)用有協(xié)同作用，并發(fā)現(xiàn)納米雄黃誘導(dǎo)腫瘤人皮膚鱗癌A431細(xì)胞凋亡的機(jī)理與上調(diào)P53的表達(dá)和下調(diào)Bcl-2的表達(dá)有關(guān)。結(jié)論:納米雄黃可有效誘導(dǎo)皮膚鱗癌A431細(xì)胞發(fā)生凋亡。納米雄黃可以通過(guò)誘導(dǎo)腫瘤細(xì)胞凋亡等途徑實(shí)現(xiàn)抗腫瘤作用，與順鉑聯(lián)合用藥，可產(chǎn)生協(xié)同作用。
[Abstract]:Objective: to investigate the effect of nanometer realgar on apoptosis of skin squamous cell carcinoma A431 cells. To explore the mechanism of inducing apoptosis and inhibiting proliferation of tumor cells by nano-realgar. Methods: this experiment mainly includes the morphological observation of the preparation of A431 cells by mechanical preparation of nano-realgar. The mechanism of nano-realgar induced apoptosis and inhibition of proliferation of human cutaneous squamous cell carcinoma A431 cells was studied by immunohistochemistry and RT-PCR. Results: nano realgar could induce apoptosis of human skin squamous cell carcinoma A431 cell line. And synergistic effect with cisplatin. It was also found that the mechanism of apoptosis induced by nano-realgar was related to the up-regulation of p53 expression and down-regulation of Bcl-2 expression. Conclusion: Nano-realgar can effectively induce apoptosis of human skin squamous cell carcinoma A431 cells. The anti-tumor effect was achieved by inducing apoptosis of tumor cells. In combination with cisplatin, synergistic effects can be produced.
