【摘要】：目的：對皮膚腫瘤進(jìn)行中醫(yī)文獻(xiàn)復(fù)習(xí)，并通過檢測人皮膚鱗癌A431細(xì)胞用納米雄黃處理后細(xì)胞增殖抑制及血管生長調(diào)控因子表達(dá)情況，，探討納米雄黃抑制血管生長抗腫瘤的作用機(jī)理。 方法：查詢并復(fù)習(xí)中醫(yī)文獻(xiàn)資料，對皮膚鱗癌A431細(xì)胞進(jìn)行體外培養(yǎng)，按實(shí)驗(yàn)分組加藥后觀察其形態(tài)及生長狀況變化，并分別用MTT法、免疫組織化學(xué)法和RT-PCR法檢測加藥后細(xì)胞增殖抑制、色素上皮源性因子(PEDF)和VEGFmRNA表達(dá)情況，探討納米雄黃抑制皮膚鱗癌新生血管生成的作用機(jī)理。 結(jié)果：納米雄黃可以抑制人皮膚鱗癌A431細(xì)胞增殖，且與順鉑聯(lián)合應(yīng)用有協(xié)同作用，并發(fā)現(xiàn)納米雄黃抑制A431細(xì)胞增殖的作用與上調(diào)PEDF的表達(dá)和下調(diào)VEGF的表達(dá)以抑制新生血管生成有關(guān)。 結(jié)論：體外實(shí)驗(yàn)表明納米雄黃可顯著抑制人皮膚鱗癌A431細(xì)胞增殖，并通過抑制其新生血管生成達(dá)到抗腫瘤細(xì)胞生長的效果，與順鉑有協(xié)同作用。
[Abstract]:Objective: to review the literatures of traditional Chinese medicine (TCM) on skin tumors and to detect the inhibition of proliferation and the expression of vascular growth regulation factor (VEGF) in human skin squamous cell carcinoma A431 cells treated with nano-realgar. To explore the mechanism of inhibiting angiogenesis and anti-tumor by nanometer realgar. Methods: the A431 cells of cutaneous squamous cell carcinoma were cultured in vitro. The morphology and growth of A431 cells were observed by MTT. Immunohistochemical method and RT-PCR method were used to detect the expression of PEDFand VEGFmRNA, and to explore the mechanism of inhibiting angiogenesis of skin squamous cell carcinoma by nano-realgar. Results: Nano-realgar could inhibit the proliferation of human skin squamous cell carcinoma A431 cells and had synergistic effect with cisplatin. It was also found that the inhibitory effect of nano-realgar on the proliferation of A431 cells was related to the up-regulation of PEDF expression and down-regulation of VEGF expression to inhibit angiogenesis. Conclusion: nano realgar can significantly inhibit the proliferation of human skin squamous cell carcinoma A431 cells in vitro and can inhibit the growth of human skin squamous cell carcinoma A431 cells by inhibiting its neovascularization. It has synergistic effect with cisplatin.
