【摘要】：目的探討自噬在富氫液治療帶狀皰疹后神經(jīng)痛(PHN)中的作用。方法雄性SD大鼠100只,采用隨機(jī)數(shù)字表法,將其分為5組(n=20):對(duì)照組、PHN組、富氫液處理的PHN組(PHN-H2組)、富氫液和3-MA同時(shí)處理的PHN組(PHN-H2-3-MA組)、富氫液和雷帕霉素(Rap)同時(shí)處理的PHN組(PHN-H2-Rap組)。通過(guò)水痘帶狀皰疹病毒(VZV)接種建立PHN大鼠模型。造模成功的大鼠給予腹腔注射15 mg/kg 3-MA或10 mg/kg Rap,隔日1次,連續(xù)3次。富氫液處理方式為于造模后每天2次,腹腔注射富氫液10 m L/kg,連續(xù)7 d。50只大鼠分別于造模后3、7、14、21 d,測(cè)定機(jī)械縮足反應(yīng)閾(PWT);另50只大鼠,于造模后7 d,收集脊椎腰膨大處,ELISA測(cè)定腫瘤壞死因子α(TNF-α)、白細(xì)胞介素1β(IL-1β)和IL-6的含量,Western blot法測(cè)定自噬蛋白微管相關(guān)蛋白1輕鏈3、beclin 1、P62的水平。結(jié)果與對(duì)照組比較,PHN組大鼠PWT值減少,TNF-α、IL-1β、IL-6、LC3Ⅱ和beclin 1水平增加,P62含量減少;與PHN組比較,PHN-H2組和PHN-H2-Rap組大鼠PWT值增加,TNF-α、IL-1β和IL-6水平降低,LC3和beclin 1表達(dá)進(jìn)一步增加,P62含量增加;與PHN-H2組比較,PHN-H2-3-MA組大鼠PWT值明顯減少,TNF-α、IL-1β、IL-6水平增加、LC3和beclin 1水平降低、P62含量增加。結(jié)論富氫液可以通過(guò)激活自噬減輕PHN大鼠的機(jī)械痛敏和炎癥因子的釋放。
[Abstract]:Objective to investigate the role of autophagy in the treatment of postherpetic neuralgia (PHN) with hydrogen rich solution. Methods 100 male SD rats were randomly divided into 5 groups: control group (PHN group). PHN group treated with hydrogen-rich liquid, PHN group treated with 3-MA and hydrogen rich solution, PHN-H2-3-MA group treated with hydrogen rich solution, PHN-H2-Rap group of PHN group treated with hydrogen rich liquid and rapamycin at the same time. The rat model of PHN was established by inoculation of varicella zoster virus (VZV). Rats were given intraperitoneal injection of 15 mg/kg 3-MA or 10 mg/kg Rapp once every other day. For 3 consecutive times, hydrogen rich solution was treated by intraperitoneal injection of 10 mL / kg hydrogen enrichment solution twice a day after modeling, and 7 d.50 rats were injected continuously for 21 days, respectively, and the mechanical foot contraction response threshold (PWTT) was measured in 50 rats. On the 7th day after modeling, the contents of TNF- 偽 TNF- 偽 TNF- 偽, IL-1 尾 and IL-6 were measured by Elisa and the level of autophagy protein microtubule-associated protein 1 light chain 3fibeclin 1 P62 was determined by Western blot.
