【摘要】：目的研究抗菌蛋白銀屑病素(psoriasin)在T細(xì)胞分泌IL-8中的作用。方法抽取正常人和銀屑病患者外周血,提取T淋巴細(xì)胞,用ELISA的方法檢測(cè)正常人和銀屑病患者外周血中IL-8的表達(dá)變化。結(jié)果未受psoriasin刺激的正常人血T細(xì)胞不能產(chǎn)生IL-8,而在psoriasin刺激2,4,8和12h后,可見(jiàn)IL-8的表達(dá)升高,且有時(shí)間和劑量依賴性,8h時(shí),產(chǎn)生IL-8的量最多,可達(dá)492pg/mL;未受psoriasin刺激的銀屑病患者血T細(xì)胞可產(chǎn)生少量IL-8,銀屑病患者外周血在psoriasin刺激2,4,8和12h后,也見(jiàn)IL-8的表達(dá)升高,4h時(shí)的產(chǎn)生量已經(jīng)相當(dāng)于正常細(xì)胞8h的產(chǎn)生量,且8h時(shí)產(chǎn)生量最多,可達(dá)1 007.5pg/mL。結(jié)論 Psoriasin刺激銀屑病患者外周血T細(xì)胞產(chǎn)生IL-8的速度比正常人更快,劑量更多。
[Abstract]:Objective to study the role of antifungal protein psoriasis in the secretion of IL-8 by T cells. Methods T lymphocytes were extracted from the peripheral blood of normal subjects and patients with psoriasis. ELISA was used to detect the expression of IL-8 in the peripheral blood of normal subjects and psoriatic patients. Results the T cells without psoriasin stimulation could not produce IL-8, but the expression of IL-8 increased after psoriasin stimulation for 2h and 12h. The amount of IL-8 produced was the highest (492pg / mL) at 8h in a time-and dose-dependent manner, and a small amount of IL-8 was produced in the blood T cells of psoriatic patients without psoriasin stimulation, and the peripheral blood of psoriatic patients was stimulated by psoriasin for 2h and 12h. It was also found that the expression of IL-8 increased at 4 h, and the amount of IL-8 produced was the same as that of normal cells at 8 h, and at 8 h it was the highest, up to 1 007.5 PG / mL. Conclusion Psoriasin stimulates the production of IL-8 in peripheral blood T cells of psoriatic patients more rapidly than that of normal controls.
