百草益膚制劑對(duì)小鼠變應(yīng)性接觸性皮炎的影響及機(jī)制研究

【摘要】:變應(yīng)性接觸性皮炎(Allergic contact dermatitis,ACD)是皮膚或黏膜接觸致敏物后主要由T細(xì)胞介導(dǎo)的遲發(fā)型變態(tài)反應(yīng)引起的炎癥性皮肽病,包括來源于T細(xì)胞等的多種細(xì)胞因子、趨化因子以及共刺激分子參與了整個(gè)炎癥過程。目前大多數(shù)觀點(diǎn)認(rèn)為ACD是由分泌IL-2、IFN-γ的Thl細(xì)胞介導(dǎo)的,其發(fā)生是因?yàn)門hl/Th2分化失衡使其免疫紊亂。ACD的治療以去除致敏原,外用糖皮質(zhì)激素,應(yīng)用保濕劑等為主。糖皮質(zhì)激素應(yīng)用是目前控制急性期變態(tài)反應(yīng)及炎癥的主要方法,但長期應(yīng)用會(huì)造成腎上腺皮質(zhì)功能低下、繼發(fā)感染等全身副作用,還可引起皮肽萎縮、毛細(xì)血管擴(kuò)張、激素性白內(nèi)障、青光眼、顏面紅斑等,從而影響兒童的生長發(fā)育和成人內(nèi)分泌系統(tǒng),停藥后易復(fù)發(fā),久用產(chǎn)生治療抵抗的現(xiàn)象也普遍存在。因此,聯(lián)合應(yīng)用其它治療方法,尋找副作用小,并且高效的治療變應(yīng)性接觸性皮炎的制劑,盡可能減少糖皮質(zhì)激素用量,甚至停用,是目前臨床治療努力的方向。 目前,我們運(yùn)用現(xiàn)代制劑學(xué)方法自制的親諾嬰兒百草益膚制劑以苦參、庫拉索蘆薈凝膠等為主要原料,其中苦參中的苦參堿可對(duì)抗急性滲出性炎癥,具有非甾體抗炎的特性,苦參堿還具有免疫抑制作用,分子機(jī)制研究表明苦參堿對(duì)P物質(zhì)誘導(dǎo)單核細(xì)胞趨化蛋白-1(MCP-1)具有干預(yù)作用;庫拉索蘆薈含較多的黏液素,其中多糖類物質(zhì)含量高,它不僅具有免疫調(diào)節(jié)功能,還有助于細(xì)胞分裂,對(duì)皮膚的保養(yǎng)修復(fù)可起重要作用。本制劑具有清熱解毒、燥濕、收斂止癢等特點(diǎn),對(duì)濕疹皮炎具有療效好、使用方便、無明顯副作用等優(yōu)點(diǎn)。本研究通過建立小鼠變應(yīng)性接觸性皮炎模型,來探討本制劑對(duì)其的影響及可能機(jī)制。 目的 通過建立小鼠變應(yīng)性接觸性皮炎模型,觀察百草益膚制劑對(duì)其的影響并探討可能機(jī)制。 方法 本文從實(shí)驗(yàn)角度,以2,4—二硝基氟苯(DNFB)誘發(fā)建立小鼠ACD模型,外用親諾嬰兒百草益膚制劑及外用嬰兒濕疹膏、丁酸氫化可的松乳膏作對(duì)照,觀察分組給藥后小鼠耳朵的腫脹度及組織病理變化,同時(shí)用ELISA及免疫組織化學(xué)方法分別檢測(cè)不同時(shí)相各組小鼠血清及耳上皮組織中白介素-2及干擾素-γ表達(dá)水平。 結(jié)果 親諾嬰兒百草益肽膏、親諾嬰兒百草益膚水、丁酸氫化可的松乳膏及嬰兒濕疹膏組均能降低小鼠左耳腫度,下調(diào)小鼠血清及皮損中白介素-2及干擾素-Y的表達(dá),且親諾嬰兒百草益膚膏與丁酸氫化可的松乳膏組療效無明顯差別(P0.05),與親諾嬰兒百草益膚水及嬰兒濕疹膏組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論 百草益肽制劑對(duì)小鼠變應(yīng)性接觸性皮炎有抑制作用,其作用機(jī)制可能與抑制白介素-2及干擾素-γ等細(xì)胞因子的釋放有關(guān)。 意義 本研究為該藥的開發(fā)及臨床應(yīng)用提供了實(shí)驗(yàn)依據(jù),從而印證該藥治療ACD及此類疾病如濕疹等的可行性,對(duì)解除患者痛苦,提高患者生活質(zhì)量,充分發(fā)揮中醫(yī)中藥的優(yōu)勢(shì)具有重要意義。
[Abstract]:Allergic contact dermatitis (Allergic contact, dermatitis, ACD) is caused by delayed hypersensitivity skin or mucosal contact allergens was mainly mediated by T cells in the inflammatory skin disease, including a variety of cytokines from T cells, chemokines and costimulatory molecules involved in the inflammatory process at present the majority view ACD is secreted by IL-2 cells, Thl IFN- gamma guide, it occurs when the Thl/Th2 imbalance in the treatment of the immune disorder.ACD to remove allergens, topical corticosteroids, application of moisturizing agent. Glucocorticoid application is the main method to control allergy and the acute phase of inflammation, but long-term use will cause adrenal insufficiency, systemic side effects of secondary infection, can also cause skin atrophy, telangiectasia, steroid induced cataract, glaucoma, facial red spots, thus. Sound growth and endocrine system of adult children, easy to relapse after drug withdrawal, long produced treatment resistance phenomenon is also widespread. Therefore, combined with other treatments for side effects, preparation and efficient treatment of allergic contact dermatitis, reducing dosage of glucocorticoid and even disabled is the clinical treatment direction.
At present, we made use of modern preparation methodology Pro Nobel herbal preparations for the baby skin of Sophora flavescens, Kuraso aloe gel as the main raw materials, wherein the Matrine in matrine against acute exudative inflammation, has the characteristics of non steroidal anti-inflammatory, matrine has immunosuppressive effect, the molecular mechanism study indicated that matrine on substance P induced monocyte chemoattractant protein -1 (MCP-1) with intervention; Kuraso aloe contains more mucus, the polysaccharide content is high, it not only has the immunoregulatory function, also contributes to cell division, which plays an important role in the repair and maintenance of the skin. The preparation has heat clearing and detoxifying, removing dampness. The convergence characteristics of dermatitis and eczema itching, has good curative effect, convenient use and has no obvious side effects. In this study we established a mouse model of allergic contact dermatitis, to investigate the effect of the preparation of the And possible mechanisms.
objective
Through the establishment of a model of allergic contact dermatitis in mice, the effect of a hundred herbs and beneficial skin preparation on the dermatitis was observed and the possible mechanism was discussed.
Method
This article from the experimental point of view, the 2,4 - two 4-dinitrofluorobenzene (DNFB) induced mice ACD model, topical herbal preparation Pro Nobel baby skin and topical baby eczema cream, Hydrocortisone Cream butyric acid group, observation group after administration of the swelling of mouse ear and pathological changes, the expression level at the same time with ELISA and immunohistochemistry methods were detected by the mice serum and ear epithelial tissues of interleukin -2 and interferon gamma.
Result
Pro Nobel baby herbal peptide cream, pro Nobel herbal baby lotion, baby eczema cream and Hydrocortisone Cream butyric acid group could reduce the mice left ear swelling, down regulated expression of interleukin in serum and skin lesions in mice -2 and interferon -Y, and pro Nobel baby herbal skin cream and butyric acid groups had no significant difference (Hydrocortisone Cream P0.05), and pro Nobel baby herbal toner and baby eczema cream group, the difference was statistically significant (P0.05).
conclusion
Paraquat peptide preparation can inhibit allergic contact dermatitis in mice, and its mechanism may be related to the release of cytokines such as interleukin -2 and interferon gamma.
Significance
This study provides experimental evidence for the development and clinical application of the drug, so as to confirm the feasibility of the drug in the treatment of ACD and other diseases such as eczema. It is important for relieving the pain of patients, improving the quality of life of patients and giving full play to the advantages of traditional Chinese Medicine.
