散血草對血管瘤裸鼠模型瘤體生長的影響及其機(jī)制探討

【摘要】:目的:通過建立血管瘤裸鼠模型,觀察散血草對血管瘤裸鼠模型生長的影響并探討其可能的作用機(jī)制。方法:采用組織塊移植法,在無菌條件下取嬰幼兒增生期毛細(xì)血管瘤手術(shù)標(biāo)本切成小塊,植入裸鼠皮下,制成嬰幼兒血管瘤裸鼠異種移植動(dòng)物模型。移植60天后,取移植成功的實(shí)驗(yàn)動(dòng)物模型建立隨機(jī)對照實(shí)驗(yàn),分為4個(gè)實(shí)驗(yàn)組,分別給予以下不同干預(yù)措施:散血草組在血管瘤瘤體表面外用自制的散血草搽劑每日一次;激素組在血管瘤瘤體內(nèi)均勻注射曲安奈德注射液0.05ml(4mg/ml)一次;散血草加激素組瘤體表面外用散血草搽劑,并在瘤體內(nèi)注射曲安奈德注射液0.05ml(4mg/ml)一次;對照組瘤體表面外用對照物每日一次。給予不同干預(yù)措施后,,通過對血管瘤裸鼠模型瘤體體積的測量,監(jiān)測血管瘤在裸鼠模型體內(nèi)的生長情況;用藥后15天處死實(shí)驗(yàn)裸鼠模型,取出血管瘤組織制成切片,HE染色進(jìn)行組織學(xué)分析,并作免疫組織化學(xué)(S-P)染色檢測HIF-1α、bcl-2和Ki-67表達(dá)的陽性指數(shù),分析血管瘤在裸鼠模型體內(nèi)的增殖和凋亡情況。使用SPSS14.0統(tǒng)計(jì)軟件對所有實(shí)驗(yàn)結(jié)果進(jìn)行統(tǒng)計(jì)學(xué)分析,以P0.05為有顯著性差異。結(jié)果:制作的血管瘤裸鼠模型生長良好,血管瘤瘤體體積大小均勻,經(jīng)對照物、散血草、散血草加激素、激素處理15天后,對照組裸鼠模型血管瘤瘤體體積繼續(xù)增大,標(biāo)本大體及鏡下觀測血管瘤瘤體呈增生期改變;散血草組、激素組、散血草加激素組裸鼠模型血管瘤瘤體體積與對照組相比均有縮小,各組之間血管瘤瘤體體積也均有顯著性差異,組織標(biāo)本觀察到血管減少,瘤體萎縮、退化,血管腔閉塞、破裂,血管間纖維脂肪組織浸潤,其中散血草加激素組最明顯,激素組次之,散血草組血管瘤瘤體縮小程度最小。散血草組、激素組、散血草加激素組HIF-1α、bcl-2和Ki-67陽性指數(shù)與對照組相比均有減少,而且各組間相比也均有顯著性差異,HIF-1α、bcl-2和Ki-67陽性指數(shù)在散血草加激素組減少最明顯,激素組次之,散血草組瘤體減少程度最小。結(jié)論:散血草外用可以抑制血管瘤裸鼠移植模型瘤體的增殖;散血草外用與糖皮質(zhì)激素(曲安奈德)聯(lián)合應(yīng)用,對血管瘤裸鼠移植模型瘤體增殖的抑制作用強(qiáng)于單用散血草或糖皮質(zhì)激素;單用散血草對血管瘤裸鼠移植模型瘤體增殖的抑制作用不及單用糖皮質(zhì)激素;散血草外用對血管瘤裸鼠移植模型瘤體增殖的抑制作用,可能與下調(diào)HIF-1α、bcl-2和Ki-67的表達(dá)水平有關(guān)。
[Abstract]:Objective: to establish a nude mouse model of hemangioma, to observe the effect of Sanguisegrass on the growth of nude mice with hemangioma and to explore its possible mechanism. Methods: tissue mass transplantation was used. In the aseptic condition, the capillary hemangioma in the proliferative phase of infant was cut into small pieces and implanted into the subcutaneous of nude mice to make the xenotransplantation animal model of infantile hemangioma. After 60 days of transplantation, the xenotransplantation model was made. The successful experimental animal model was used to establish a randomized controlled experiment, which was divided into four experimental groups, and were given the following different intervention measures: the hemangioma group was treated with self-made Sanguisegrass liniment once a day on the surface of the hemangioma; In the hormone group, triamcinolone acetonide injection (0.05ml / ml) was injected evenly into the hemangioma. In the control group, the tumor surface was treated with Xanthoxylum liniment and triamcinolone acetonide injection (0.05 mg / ml) was injected into the tumor body. After different intervention measures were given, the growth of hemangioma in nude mice was monitored by measuring the volume of tumor in nude mice. 15 days after administration, the nude mice were killed, the hemangioma tissue was taken out and stained with HE for histological analysis, and HIF-1 偽 was detected by immunohistochemical staining. The positive index of bcl-2 and Ki-67 expression was used to analyze the proliferation and apoptosis of hemangioma in nude mice model. SPSS14.0 statistical software was used to analyze all the experimental results. Results: the model of hemangioma in nude mice grew well and the volume of hemangioma tumor was uniform. In the control group, the tumor volume of hemangioma in nude mice model continued to increase, and the changes of hemangioma tumor were observed under microscope and gross. Compared with the control group, the tumor volume of the hemangioma model of the control group was reduced, and the volume of the hemangioma tumor was significantly different among the three groups. The vasculization was observed in the tissue specimen. Tumor atrophy, degeneration, lumen occlusion, rupture, intervascular fibrous adipose tissue infiltration, in which the most obvious group, hormone group, hormone group, the smallest degree of reduction of hemangioma tumor. The positive index of HIF-1 偽 bcl-2 and Ki-67 in the hormone group and SXC plus hormone group were all decreased compared with the control group, and there was a significant difference in HIF-1 偽 between the two groups. The positive index of bcl-2 and Ki-67 decreased most obviously in the group of Sanguisuegrass plus hormone, followed by the group of hormone. Conclusion: the external application of Sanguisegrass can inhibit the proliferation of tumor in nude mice model of hemangioma transplantation. In combination with triamcinolone acetonide (triamcinolone acetonide), the inhibition of tumor proliferation in nude mice with hemangioma transplantation was stronger than that of single hemangioma grafts or glucocorticoids. The inhibitory effect of SXC on tumor proliferation in nude mice was not as good as that of glucocorticoid alone. The inhibitory effect of Sanguisegrass on the proliferation of hemangioma xenografts in nude mice may be related to the down-regulation of the expression of HIF-1 偽 bcl-2 and Ki-67.
