大鼠急性甲醇中毒大腦皮質(zhì)單核細胞趨化蛋白-1蛋白的表達

【摘要】:目的觀察大鼠急性甲醇中毒后單核細胞趨化蛋白1(MCP-1)在大腦皮質(zhì)的表達變化。方法應(yīng)用通有混合氣體(N2O/O2)的密閉有機玻璃箱并灌胃相應(yīng)劑量甲醇溶液復(fù)制急性高劑量和低劑量甲醇中毒大鼠模型,運用Western Blot和免疫組化染色檢測不同時間點大腦皮質(zhì)中MCP-1的表達變化。結(jié)果 Western Blot顯示急性甲醇中毒后低劑量組和高劑量組MCP-1蛋白表達在12 h明顯升高(P0.01),24 h達高峰,然后逐漸下降,3 d仍高于對照組(P0.01)。高劑量組MCP-1蛋白表達在12 h和24 h高于低劑量組(P0.05),3 d組MCP-1的表達無顯著差異。免疫組化染色顯示對照組大鼠大腦未見明顯MCP-1陽性表達,急性甲醇中毒后,高劑量組24 h MCP-1陽性表達數(shù)增加,3 d達高峰,和對照組相比有顯著差異(P0.01),陽性表達多集中在梨狀皮質(zhì),1周陽性反應(yīng)減少,但與對照組相比仍有顯著差異(P0.01)。結(jié)論大鼠急性甲醇中毒后,腦組織MCP-1蛋白表達顯著增高并持續(xù)一定時間,可能參與了腦組織病理變化過程,程度與甲醇中毒劑量有關(guān)。
[Abstract]:Objective to observe the expression of monocyte chemoattractant protein 1 (MCP-1) in cerebral cortex after acute methanol poisoning in rats. Methods the rat models of acute high dose and low dose methanol poisoning were established by using closed plexiglass box with mixed gas (N2O/O2) and the corresponding dose of methanol solution. The expression of MCP-1 in cerebral cortex was detected by Western Blot and Immunohistochemical staining. Results Western Blot showed that the expression of MCP-1 protein in low dose group and high dose group increased significantly at 12 h (P 0.01), reached the peak at 24 h, and then decreased gradually, which was still higher than that in control group at 3 d (P 0.01). The expression of MCP-1 protein in high dose group was higher than that in low dose group at 12 h and 24 h (P 0.05), but there was no significant difference in MCP-1 expression in 3 d group. After acute methanol poisoning, the positive expression of MCP-1 in the high dose group increased at 24 h and reached the peak at 3 d, which was significantly different from that in the control group (P 0.01). The positive expression was mostly concentrated in the piriform cortex and decreased in one week, but there was still significant difference between the high dose group and the control group (P 0.01). Conclusion the expression of MCP-1 protein in brain tissue of rats with acute methanol poisoning is significantly increased and lasts for a certain period of time, which may be involved in the pathological changes of brain tissue, and the degree is related to the dose of methanol poisoning.
