金黃色葡萄球菌顯著增加系統(tǒng)性紅斑狼瘡患者中性粒細(xì)胞NETs的產(chǎn)生

【摘要】:目的探討金黃色葡萄球菌對(duì)系統(tǒng)性紅斑狼瘡(SLE)中性粒細(xì)胞胞外誘捕網(wǎng)(NETs)產(chǎn)生的影響及意義。方法收集SLE穩(wěn)定者22例、活動(dòng)者20例和健康對(duì)照18例,分離中性粒細(xì)胞并加金黃色葡萄球菌共培養(yǎng),檢測(cè)金黃色葡萄球菌刺激后NETs的主要成分,DNA試劑盒提取測(cè)定游離DNA(f DNA)及酶聯(lián)免疫吸附測(cè)定(ELISA)法測(cè)上清LL-37,并分析其與SLE臨床及實(shí)驗(yàn)指標(biāo)的關(guān)系。結(jié)果(1)SLE穩(wěn)定組、活動(dòng)組、健康對(duì)照組3組中性粒細(xì)胞培養(yǎng)上清f DNA量3組間差異有統(tǒng)計(jì)學(xué)意義,SLE穩(wěn)定組高于活動(dòng)組和健康對(duì)照組(P分別是0.008、0.027),SLE活動(dòng)組和健康對(duì)照組差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);LL-37比較無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。(2)加入金黃色葡萄球菌共培養(yǎng),培養(yǎng)上清f DNA量的比較差異有統(tǒng)計(jì)學(xué)意義,SLE活動(dòng)組、穩(wěn)定組均高于健康對(duì)照組(P0.05),SLE活動(dòng)組與穩(wěn)定組無(wú)顯著性差異(P0.05);LL-37比較無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。(3)加入金黃色葡萄球菌共培養(yǎng)后,3組f DNA及LL-37量均較刺激前顯著增加,差異均有統(tǒng)計(jì)學(xué)意義(f DNA P分別是0.000、0.000、0.001,LL-37 P均為0.000);培養(yǎng)前后f DNA的差值SLE活動(dòng)組顯著高于穩(wěn)定組和健康對(duì)照組(P0.05),SLE穩(wěn)定組、健康對(duì)照組差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);LL-37差值組間比較均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論金黃色葡萄球菌刺激后,SLE患者NETs產(chǎn)生/釋放量顯著增加,SLE活動(dòng)者最顯著,金黃色葡萄球菌對(duì)SLE疾病活動(dòng)可能有影響。
[Abstract]:Objective To study the effect and significance of S. aureus on the extracellular trapping net (NEs) of systemic lupus erythematosus (SLE). Methods 22 cases of SLE,20 cases of active and 18 healthy controls were collected, and the main components of NEs after stimulation of S. aureus were detected by co-culture with Staphylococcus aureus. Serum free DNA (f DNA) and enzyme-linked immunosorbent assay (ELISA) were used to determine the relationship between the clinical and experimental indexes of SLE. Results (1) There was no significant difference between the three groups in the control group, the group of active SLE, the control group and the control group (P = 0.008, 0.027), and there was no significant difference between the SLE group and the healthy control group (P <0.05). LL-37 was not statistically significant (P0.05). (2) The results showed that there was no significant difference between the activity of SLE and the stable group (P <0.05), and there was no significant difference between the SLE and the stable group (P0.05). (3) After the co-culture of staphylococcus aureus, the amount of f-DNA and LL-37 was significantly increased in 3 groups (0.000, 0.000, 0.001, LL-37P was 0.000), and the difference of the difference of f DNA before and after culture was significantly higher than that of stable group and healthy control group (P0.05). There was no significant difference between the control group and the control group (P0.05). Conclusion After the stimulation of S. aureus, the generation/ release of the patients with SLE is significantly increased, and the most significant in the patients with SLE, and the effect of S. aureus on the activities of SLE may be affected.
