2型糖尿病頸動脈粥樣硬化患者血清腦源性神經(jīng)營養(yǎng)因子與腫瘤壞死因子-α水平相關(guān)性研究

【摘要】:目的:通過研究合并頸動脈粥樣硬化的2型糖尿病患者血清腦源性神經(jīng)營養(yǎng)因子(BDNF)、腫瘤壞死因子-α (TNF-α)及外周血BDNF mRNA表達水平的變化,探討2型糖尿病患者中BDNF、TNF-α與頸動脈粥樣硬化的關(guān)系及可能機制,以及BDNF與TNF-α的相關(guān)性。方法:選取入住我院的2型糖尿病患者100例,根據(jù)彩色多普勒超聲檢查實驗對象頸動脈內(nèi)中膜層厚度將其分為合并頸動脈粥樣硬化患者52例,無頸動脈粥樣硬化患者48例,并另外選取50例健康者作為對照組,檢查實驗對象頸動脈內(nèi)中膜層厚度、血脂、空腹血糖、空腹胰島功能、糖化血紅蛋白,并檢測TNF-α、BDNF濃度及外周血BDNF mRNA表達水平;使用單因素方差分析對3組間均數(shù)進行比較,使用獨立樣本t檢驗對2組間進行比較,使用Pearson相關(guān)分析方法分析2型糖尿病合并頸動脈粥樣硬化患者血清BDNF與TNF-α相關(guān)性。結(jié)果:2型糖尿病伴頸動脈粥樣硬化組患者血清BDNF濃度、BDNF mRNA表達水平明顯低于單純2型糖尿病組及正常對照組(P0.05)。2型糖尿病伴頸動脈粥樣硬化組患者血清TNF-α濃度明顯高于單純糖尿病組及正常對照組(P0.05)。2型糖尿病合并頸動脈粥樣硬化組患者血清BDNF與TNF-α水平相關(guān)(r=-0.378,P0.05)。結(jié)論:血清TNF-α與BDNF水平相關(guān),提示TNF-α與BDNF共同參與了2型糖尿病頸動脈粥樣硬化的發(fā)病。TNF-a in patientS with type 2 diabeteS mellituS and

To investigate the relationship between BDNF,TNF- 偽 and carotid atherosclerosis in type 2 diabetic patients and its possible mechanism, and the correlation between BDNF and TNF- 偽. Methods: 100 patients with type 2 diabetes mellitus admitted to our hospital were divided into 52 patients with carotid atherosclerosis and 48 patients without carotid atherosclerosis according to the thickness of carotid intima media. In addition, 50 healthy subjects were selected as control group. Carotid intima-media thickness, blood lipid, fasting blood glucose, fasting islet function, glycosylated hemoglobin, TNF- 偽, BDNF concentration and BDNF mRNA expression in peripheral blood were measured. Single factor analysis of variance (ANOVA) was used to compare the mean of the three groups, independent sample t-test was used to compare the two groups, and Pearson correlation analysis was used to analyze the correlation between serum BDNF and TNF- 偽 in patients with type 2 diabetes mellitus and carotid atherosclerosis. Results: the serum BDNF concentration in patients with type 2 diabetes mellitus with carotid atherosclerosis was determined. The level of BDNF mRNA expression was significantly lower in type 2 diabetes mellitus group than in normal control group (P0.05). The serum TNF- 偽 level in type 2 diabetes patients with carotid artery atherosclerosis was significantly higher than that in simple diabetes mellitus group and normal control group (P0.05). P0.05).
