西格列汀對(duì)高糖下系膜細(xì)胞細(xì)胞外基質(zhì)表達(dá)的影響

【摘要】:目的探討高糖對(duì)腎小球系膜細(xì)胞細(xì)胞外基質(zhì)表達(dá)的影響,以及西格列汀(SIT)對(duì)其的干預(yù)作用。方法培養(yǎng)腎小球系膜細(xì)胞,分為對(duì)照(Con)組、高糖(HG)組,以及高糖聯(lián)合不同濃度的SIT組(HG+S1組、HG+S5組、HG+S10組和HG+S20組)。培養(yǎng)24 h、48 h后,用MTT法測(cè)定細(xì)胞增殖程度,ELISA檢測(cè)細(xì)胞培養(yǎng)上清層粘連蛋白(LN)、大鼠Ⅳ型膠原蛋白(Col-Ⅳ)、轉(zhuǎn)化生長因子β1(TGF-β1)蛋白表達(dá)的變化。結(jié)果高糖培養(yǎng)24 h、48 h后,可觀察到不同濃度的SIT均可抑制高糖誘導(dǎo)的系膜細(xì)胞增殖;SIT組與HG組比較,差異有統(tǒng)計(jì)學(xué)意義;Con組腎小球系膜細(xì)胞可分泌一定量的LN、Col-Ⅳ和TGF-β1,高糖刺激后LN、Col-Ⅳ和TGF-β1分泌增加,而SIT對(duì)系膜細(xì)胞分泌物的抑制隨其濃度的升高而增強(qiáng)。結(jié)論SIT可抑制高糖條件下系膜細(xì)胞的增殖,且能降低高糖條件下LN、Col-Ⅳ、TGF-β1的表達(dá),SIT可能對(duì)糖尿病患者的腎臟有一定的保護(hù)作用。
