【摘要】：目的檢測結締組織生長因子(connectivetissue growth factor,CTGF)在類風濕關節(jié)炎(rheumatoid arthritis,RA)患者關節(jié)滑液及滑膜中的表達,探索CTGF在RA關節(jié)病變中可能發(fā)揮的作用。方法采用酶聯(lián)免疫法(enzyme-linked imm-unosorbent assay,ELISA)方法檢測中國醫(yī)科大學附屬第一醫(yī)院2013年1月至12月收治的10例RA患者和8例骨關節(jié)炎(osteoar thritis,OA)患者關節(jié)滑液CTGF水平;免疫組化法檢測3例RA患者和2例OA患者關節(jié)滑膜CTGF的表達;Real-time PCR檢測向體外培養(yǎng)的成纖維樣滑膜細胞(FLS)中加入外源性腫瘤壞死因子α(tumor necrosis factor alpha,TNF-α)后對CTGF表達的影響。結果 RA組的關節(jié)滑液CTGF水平高于OA組[(21.00±10.20)μg/L對(8.53±7.71)μg/L)],差異有統(tǒng)計學意義(P0.05);CTGF在RA滑膜組織主要表達于滑膜襯里細胞及襯里下層細胞,而OA滑膜組織僅少量CTGF表達;TNF-α可以上調FLS表達CTGF。結論 RA患者滑液CTGF水平升高,滑膜組織CTGF高表達,提示CTGF可能參與了RA關節(jié)炎癥的發(fā)生發(fā)展。TNF-α上調FLS的CTGF表達,這可能是導致RA關節(jié)病變的重要因素之一。
[Abstract]:Objective to detect the expression of connective tissue growth factor (connectivetissue growth factor,CTGF) in synovial fluid and synovium of patients with rheumatoid arthritis (rheumatoid arthritis,RA) and to explore the role of CTGF in RA joint lesions. Methods the levels of CTGF in synovial fluid of 10 patients with RA and 8 patients with osteoarthritis (osteoar thritis,OA) were detected by enzyme linked immunosorbent assay (enzyme-linked imm-unosorbent assay,ELISA) from January to December 2013 in the first affiliated Hospital of China Medical University. The expression of CTGF in synovial membrane of 3 patients with RA and 2 patients with OA was detected by immunohistochemistry. Real-time PCR was used to detect the effect of exogenous tumor necrosis factor 偽 (tumor necrosis factor alpha,TNF- 偽 (TNF- 偽) on the expression of CTGF in cultured fibroblast (FLS). Results the level of CTGF in synovial fluid in RA group was significantly higher than that in OA group [(21.00 鹵10.20) 渭 g / L vs (8.53 鹵7.71) 渭 g / L] (P0.05). CTGF was mainly expressed in synovial lining cells and sublining cells of RA, but only a small amount of CTGF was expressed in OA synovium, and TNF- 偽 could up-regulate FLS expression of CTGF.. Conclusion the increase of CTGF level in synovial fluid and the high expression of CTGF in synovial tissue in patients with RA suggest that CTGF may be involved in the development of RA arthritis. TNF- 偽 upregulated CTGF expression of FLS, which may be one of the important factors leading to RA joint lesion.
