1型糖尿病視網(wǎng)膜病變大鼠脂聯(lián)素水平與TGF-β1及ICAM-1的關(guān)系

【摘要】：目的:糖尿病視網(wǎng)膜病變(DR)是其并發(fā)癥中最為常見和嚴(yán)重的,是成人致盲的疾病之一。世界范圍內(nèi)糖尿病患者人數(shù)的不斷增大,使DR成為了視力喪失以及視功能損害的主要原因。研究脂聯(lián)素(ADPN)與l型糖尿病大鼠模型的視網(wǎng)膜組織中轉(zhuǎn)化生長因子-β1及細(xì)胞間粘附分子-1之間的關(guān)系,以及各個因子分別與DR的關(guān)系。方法:選取清潔級、近交SD(Sprague Dawley)雄性大鼠40只(體重介于180g-220g)分為2組,正常組(NC組,n=16),糖尿病組(DM組,n=24)。DM組的大鼠給予腹腔注射STZ(按計(jì)量為65mg/kg)一次足量給藥,注射藥物STZ 72h后測DM組大鼠血清Glu≥16.7mmol/L,出現(xiàn)尿頻、飲水及食量增多的癥狀,即大鼠T1DM模型制作成功。NC組大鼠用等計(jì)量的枸櫞酸鉀的緩沖溶液,腹腔一次性給藥。2組大鼠均給予一般飲食,于相同的情況下觀察7日。模型組始終滿足1型糖尿病(T1DM)模型的條件,未見轉(zhuǎn)復(fù)正常的納入實(shí)驗(yàn)組。后2組大鼠均繼續(xù)給予普通飼料喂養(yǎng),分別觀察4周及8周時2組大鼠視網(wǎng)膜病變的程度、組織中TGF-β1、ICAM-l及血清中ADPN的濃度。根據(jù)實(shí)驗(yàn)結(jié)果以及監(jiān)測數(shù)據(jù)的分析和比較,討論它們的關(guān)系。結(jié)果:l、T1DM大鼠的血清中ADPN含量的水平明顯較正常時降低,且隨著病程時間的延長ADPN的水平越低。2、視網(wǎng)膜組織中的轉(zhuǎn)化生長因子-β1及細(xì)胞間粘附分子-1的表達(dá)隨著糖尿病病程延長,組織間表達(dá)明顯增加。3、視網(wǎng)膜組織病變的嚴(yán)重程度是伴隨T1DM病程的延長,逐漸加重的。結(jié)論:1型糖尿病大鼠的血清脂聯(lián)素水平是隨著病程的發(fā)展而逐漸減低的,并且與正常組比較其水平是明顯下降的。視網(wǎng)膜組織中TGF-β1及ICAM-1的表達(dá)隨病程的進(jìn)展是明顯升高的,纖維化程度明顯加重�？烧J(rèn)為隨著脂聯(lián)素的減少TGF-β1及細(xì)胞間粘附分子-1的表達(dá)逐漸增加,視網(wǎng)膜病變程度更高。
[Abstract]:Objective: diabetic retinopathy (DR) is one of the most common and severe complications of diabetic retinopathy. With the increasing number of diabetic patients worldwide, DR has become the main cause of visual loss and visual impairment. To study the relationship between adiponectin (ADPN) and transforming growth factor- 尾 1 (TGF- 尾 1) and intercellular adhesion molecule-1 (ICAM-1) in retina of type 1 diabetic rats, and the relationship between each factor and DR. Methods: forty inbred SD (Sprague Dawley) male rats (weight between 180g-220g and clean grade) were divided into two groups: normal group (NC group, nong16), diabetic group (DM group), and normal control group (NC group, n = 16). Rats in 24). DM group were given intraperitoneal injection of STZ (measured as 65mg/kg) in one dose. The serum Glu of DM group was measured 72 hours after the injection of STZ. The rats in DM group showed symptoms of frequent urination, drinking water and increasing food intake. Rats in NC group were given intraperitoneal administration of potassium citrate buffer solution. The rats in both groups were given general diet and observed for 7 days under the same conditions. The model group always satisfied the condition of type 1 diabetes mellitus (T1DM) model and did not return to normal in the experimental group. Rats in the latter two groups were continued to be fed with common diet. The degree of retinopathy, the concentration of TGF- 尾 1-ICAM-l in tissues and the concentration of ADPN in serum of the two groups were observed at 4 and 8 weeks, respectively. According to the experimental results and the analysis and comparison of the monitoring data, the relationship between them is discussed. Results: the level of ADPN in serum of T1DM rats was significantly lower than that of normal rats, and the lower the level of ADPN was with the prolongation of the course of disease. The expression of transforming growth factor- 尾 1 and intercellular adhesion molecule-1 in retinal tissues increased significantly with the prolongation of the course of diabetes mellitus. The severity of retinopathy was accompanied by the prolongation of the course of T1DM. Gradually aggravating. Conclusion: the serum adiponectin level of type 1 diabetic rats decreased gradually with the development of the course of disease, and the level of adiponectin decreased significantly compared with the normal group. The expression of TGF- 尾 1 and ICAM-1 in retinal tissue was significantly increased with the progression of the disease, and the degree of fibrosis was significantly aggravated. It can be concluded that with the decrease of adiponectin, the expression of TGF- 尾 1 and intercellular adhesion molecule-1 (ICAM-1) increases gradually, and the degree of retinopathy is higher.
