HSPB1(HSP27)表達水平與糖尿病病變程度相關性研究

【摘要】:目的:糖尿病是最常見的慢性病之一,流行率正逐漸上升。目前糖尿病的發(fā)病機制尚不明確。高血糖狀態(tài)引起的氧化應激增加是引起胰島細胞損傷及糖尿病并發(fā)癥靶器官損害的重要機制。氧化反應亢進會引起細胞凋亡,而熱休克蛋白基因HSPB1表達的HSP27蛋白是近期發(fā)現(xiàn)的介導細胞凋亡的相關分子。我們的目的是調查正常葡萄糖耐量(NGT),葡萄糖耐量異常(IGT)和2型糖尿病患者與HSP27表達水平有無相關性。方法:入選從2015年10月至2016年12月之間于寧夏醫(yī)科大學總醫(yī)院及心腦血管病醫(yī)院就診的2型糖尿病患者36例,糖耐量異;颊28例,及同期健康體檢患者22例,并且匹配年齡和性別。收集研究對象資料,包括性別、年齡、體重指數(shù)、病程、血壓、空腹血糖(FBG)、血脂、糖化血紅蛋白、糖尿病組病程等。且通過ELISA法測量每個研究對象HSP27的水平做統(tǒng)計學分析。結果:糖尿病組HSP27水平(299.33+58.92pg/ml)顯著低于IGT患者(347.46+66.24pg/ml,P0.05),高于正常對照組(211.86+70.08pg/ml,P0.05)。結論:1.人血清HSP27的表達水平與糖尿病及糖尿病發(fā)展程度有關。2.HbA1c與HSP27水平存在相關性。
[Abstract]:Objective: diabetes is one of the most common chronic diseases and the prevalence rate is increasing gradually. At present, the pathogenesis of diabetes is not clear. The increase of oxidative stress induced by hyperglycemia is an important mechanism of islet cell injury and diabetic complication target organ damage. Hyperoxidation can induce apoptosis, and the HSP27 protein expressed by heat shock protein gene (HSPB1) is a recently discovered molecule involved in cell apoptosis. Our aim was to investigate the correlation between normal glucose tolerance (NGT), impaired glucose tolerance (IGT) and type 2 diabetes mellitus (type 2 diabetes mellitus) with HSP27 expression. Methods: from October 2015 to December 2016, 36 patients with type 2 diabetes mellitus, 28 patients with impaired glucose tolerance and 22 patients with physical examination were enrolled from the General Hospital of Ningxia Medical University and the Hospital for Cardiovascular and Cerebrovascular Diseases. And match age and gender. Data were collected, including sex, age, body mass index, course of disease, blood pressure, fasting blood glucose, (FBG), glycosylated hemoglobin, course of diabetes. The level of HSP27 was measured by ELISA method. Results: the level of HSP27 in diabetic group (299.33 58.92pg/ml) was significantly lower than that in IGT group (347.46 66.24 PG / ml P 0.05) and higher than that in normal control group (211.86 70.08 PG / ml P 0.05). Conclusion 1. The expression of HSP27 in human serum was related to the degree of diabetes mellitus and the development of diabetes mellitus. 2. HbA1c was correlated with the level of HSP27.
