【摘要】：目的探討膜聯(lián)蛋白A1(Annexin A1)對(duì)糖尿病大鼠心功能及腫瘤壞死因子α(TNF-α)、白細(xì)胞介素1β(IL-1β)的影響。方法將24只SD大鼠隨機(jī)均分為正常對(duì)照組、糖尿病組;利用高糖高脂飲食飼養(yǎng)及較小劑量鏈脲佐菌素(30 mg/kg)誘導(dǎo)造成2型糖尿病模型,2組分別予以普通喂食、高糖高脂喂食;于第8周末用彩色多普勒超聲檢測(cè)左心室舒張末期容積(LVEDV)、左心室收縮末期容積(LVESV)、二尖瓣舒張?jiān)缙诜逯邓俣?二尖瓣舒張晚期峰值速度(e/a)比值、左心室射血分?jǐn)?shù)(LVEF)、每搏射血量(SV);第0、4、8周采用實(shí)時(shí)PCR測(cè)定血中Annexin A1表達(dá),采用酶聯(lián)免疫分析法測(cè)定血中TNF-α、IL-1β。結(jié)果與正常對(duì)照組對(duì)比,糖尿病組LVEDV、e/a、SV明顯下降,Annexin A1明顯上升(P0.05),8周后糖尿病組Annexin A1顯著下降(P0.01);與正常對(duì)照組對(duì)比,糖尿病組TNF-α、IL-1β上升明顯(P0.05),8周后上升更明顯(P0.01);糖尿病組Annexin A1與TNF-α、IL-1β負(fù)相關(guān)(r分別為-0.73、-0.71,P0.01)。結(jié)論 Annexin A1可改善糖尿病大鼠心功能,Annexin A1改善糖尿病大鼠心功能可能與炎癥反應(yīng)有關(guān)。
[Abstract]:Objective to investigate the effects of Annexin A1 on cardiac function and tumor necrosis factor 偽 (TNF- 偽) and interleukin 1 尾 (IL-1 尾) in diabetic rats. Methods Twenty-four Sprague-Dawley rats were randomly divided into normal control group and diabetic group, fed with high glucose and high fat diet and low dose streptozotocin (30 mg/kg). At the end of the 8th week, left ventricular end-diastolic volume (LVEDV), left ventricular end-systolic volume (LVEDV), early diastolic peak velocity (LVESV),) / end-diastolic peak velocity (E / a) ratio of left ventricular end diastolic volume (LVEDV),) were measured by color Doppler ultrasound. Left ventricular ejection fraction (LVEF),) was used to detect the expression of Annexin A1 in blood by real-time PCR and TNF- 偽 and IL-1 尾 by enzyme linked immunosorbent assay (Elisa). Results compared with normal control group, Annexin A1 decreased significantly in diabetic group (P0.05), and decreased significantly in diabetic group (P0.01), and decreased significantly in diabetic group (P0.05), and decreased significantly in diabetic group (P0.01) after 8 weeks, compared with normal control group. In diabetic group, the level of TNF- 偽 and IL-1 尾 increased significantly (P0.05) after 8 weeks (P0.01), and the correlation between TNF- 偽 IL-1 尾 and Annexin A1 was negative (r = -0.73 鹵-0.71 P0.01, respectively). Conclusion Annexin A1 can improve cardiac function in diabetic rats.
