類風(fēng)濕性關(guān)節(jié)炎患者嗜堿性粒細(xì)胞活化機(jī)制研究

【摘要】:目的:探討類風(fēng)濕性關(guān)節(jié)炎患者外周血嗜堿性粒細(xì)胞的活化情況及其機(jī)制。方法:流式檢測正常人對照組和RA患者組外周嗜堿性粒細(xì)胞活化標(biāo)記物CD203c的表達(dá)及IL-4陽性的比例;用電化學(xué)發(fā)光法檢測血清IgE水平;負(fù)選正常人嗜堿性粒細(xì)胞,以anti-IgE刺激為陽性對照,置含或不含IgE的微環(huán)境中,流式檢測CD203c的表達(dá)及IL-4陽性的比例。結(jié)果:較正常人對照組,RA患者嗜堿性粒細(xì)胞高表達(dá)CD203c和IL-4(均P0.05);RA患者血清IgE水平顯著高于正常人對照組(P0.05);RA患者血清和其血清中純化的IgE可使負(fù)選的正常人來源的嗜堿性粒細(xì)胞活化,上調(diào)CD203c表達(dá)及IL-4陽性比例。結(jié)論:RA患者嗜堿性;罨c發(fā)病相關(guān),且主要由IgE介導(dǎo),靶向嗜堿性粒細(xì)胞及其活化通路有望為RA的防治提供新的策略。
[Abstract]:Objective: to investigate the activation and mechanism of peripheral blood basophil in patients with rheumatoid arthritis. Methods: flow cytometry was used to detect the expression of peripheral basophil activation marker (CD203c) and the proportion of IL-4 positive in peripheral eosinophil activation marker in normal control group and RA group. The serum IgE level was detected by electrochemiluminescence assay. Using anti-IgE stimulation as positive control, the expression of CD203c and the proportion of IL-4 positive were detected by flow cytometry in microenvironment with or without IgE. Results: the expression of CD203c and IL-4 in serum of RA patients was significantly higher than that of normal controls (P0.05). The serum IgE levels in RA patients were significantly higher than those in normal controls (P0.05). The purified IgE in serum of RA patients and their sera could result in negative selection of eosinophils from normal people. Alkaline granulocyte activation, The expression of CD203c and the positive rate of IL-4 were upregulated. Conclusion the activation of basophil is related to the pathogenesis of RA and is mainly mediated by IgE. Targeting basophil and its activation pathway may provide a new strategy for the prevention and treatment of RA.
