頸交感神經(jīng)阻滯對(duì)糖尿病大鼠海馬P38、TLR4的影響

【摘要】:目的:觀察頸交感神經(jīng)阻滯對(duì)糖尿病大鼠海馬P38蛋白及TLR4表達(dá)的影響.方法:雄性成年大鼠64只,采用鏈佐菌(STZ)制作糖尿病大鼠模型,分為2組.A組為糖尿病+頸交感神經(jīng)阻滯組(n=32),B組為糖尿病組(n=32).水迷宮實(shí)驗(yàn)行定位航行實(shí)驗(yàn)及空間探索實(shí)驗(yàn)觀察A、B組大鼠造模前(T_1)、造模后8(T_2)、10(T_3)、12(T_4)周的認(rèn)知功能.分別于各時(shí)點(diǎn)每組各處死8只大鼠,采用Real-time RT-PCR和Western blot法測(cè)定海馬P38、TLR4的表達(dá).結(jié)果:A、B組8、10、12周逃避潛伏期較造模前明顯延長(zhǎng)、穿越平臺(tái)的次數(shù)明顯減少(P0.05).與B組比較,A組(頸交感神經(jīng)阻滯組)大鼠逃避潛伏期明顯縮短、穿越平臺(tái)的次數(shù)明顯增加(P0.05).A組各時(shí)點(diǎn)海馬P38蛋白相對(duì)表達(dá)量明顯高于B組,海馬TLR4蛋白相對(duì)表達(dá)量明顯低于于B組(P0.05).A組各時(shí)點(diǎn)海馬P38mRNA相對(duì)表達(dá)倍數(shù)明顯高于B組,海馬TLR4mRNA相對(duì)表達(dá)倍數(shù)明顯低于B組(P0.05).結(jié)論:頸交感神經(jīng)阻滯可改善糖尿病大鼠的認(rèn)知功能,可能與其降低炎癥反應(yīng)、調(diào)節(jié)P38蛋白的表達(dá)有關(guān).
[Abstract]:Aim: to observe the effect of cervical sympathetic block on the expression of P 38 protein and TLR 4 in hippocampus of diabetic rats. Methods: Sixty-four adult male rats were divided into 2 groups: group A (n = 32), group B (n = 32), and group B (n = 32) with diabetes mellitus (n = 32). Water labyrinth experiments were conducted to observe the cognitive function of rats in AHB group before and 8 (T _ 2) T _ 3 (T _ 3) ~ 12 (T _ 4) weeks after model making (T _ (1) and (T _ (2) (T _ (3) (T _ 4). Eight rats were killed in each group at each time point, and the expression of TLR4 in hippocampus was detected by Real-time RT-PCR and Western blot. Results the escape latency of 12 weeks in group B was significantly longer than that before modeling, and the frequency of crossing the platform was significantly decreased (P0.05). Compared with group B, the escape latency of rats in group A (cervical sympathetic nerve block group) was significantly shortened, and the number of times of crossing platform was significantly increased (P0.05). The relative expression of P38 protein in hippocampus of group A was significantly higher than that of group B at each time point. The relative expression of TLR4 protein in hippocampus was significantly lower than that in group B (P0.05). The relative multiple of P38 mRNA expression in hippocampus was significantly higher than that in group B at each time point (P0.05), and the relative expression multiple of TLR4 mRNA in hippocampus was significantly lower than that in group B (P0.05). Conclusion: cervical sympathetic nerve block can improve the cognitive function of diabetic rats, which may be related to the decrease of inflammation and the regulation of P38 protein expression.
