【摘要】：核膜蛋白綜合征是一類主要由細胞核膜結(jié)構(gòu)蛋白(如lamins、SUNs、emerin等)基因突變誘發(fā)的包括早衰等一系列疾病的總稱,因其多數(shù)突變發(fā)現(xiàn)于核纖層蛋白基因上,所以又被稱為核纖層蛋白綜合征,其發(fā)病機制目前還不清楚。本文以A型核纖層蛋白基因位點上的致病突變基因C1294T為切入點,研究突變對細胞核結(jié)構(gòu)與功能的影響,以及相關(guān)核膜蛋白及染色質(zhì)定位與功能的影響。研究結(jié)果表明,LMNA突變對細胞核膜的結(jié)構(gòu)與功能完整性具有影響作用,可致使細胞核膜異形甚至產(chǎn)生凸起小泡;另外還對細胞核內(nèi)染色質(zhì)的定位具有一定抑制作用,使其在細胞核內(nèi)形成不均勻的分布狀態(tài),表明該突變已經(jīng)影響到細胞核正常的結(jié)構(gòu)與功能,并可能進一步誘發(fā)相關(guān)疾病。
[Abstract]:Nuclear membrane protein syndrome (NMPS) is a class of diseases, including premature senescence, which is mainly induced by mutations in nuclear membrane structural proteins (such as lamins, SUNsSUNsOemerin, etc.), because most of the mutations are found in nuclear laminin genes. Therefore, it is also known as nuclear laminin syndrome, its pathogenesis is not clear. In this paper, the effect of mutation on nuclear structure and function and the localization and function of nuclear membrane protein and chromatin were studied by using C1294T, a pathogenic mutation gene of A type nuclear laminin gene. The results showed that LMNA mutation had an effect on the structural and functional integrity of the nuclear membrane, leading to the abnormal shape of the nuclear membrane and even the formation of bulging vesicles, and also inhibited the localization of chromatin in the nucleus. It is suggested that the mutation has affected the normal structure and function of the nucleus and may further induce related diseases.
