胰升血糖素通過環(huán)磷鳥嘌呤核苷信號通路調(diào)節(jié)MIN6細胞胰島素分泌的作用機制研究

【摘要】:目的探討不同濃度的胰升血糖素(Glg)和葡萄糖(Glu)對胰島β細胞株MIN6中胰島素(Ins)分泌功能及細胞內(nèi)環(huán)磷鳥嘌呤核苷(cGMP)信號通路的影響。方法用不同濃度Glu(0、2.8、16.7 mmol/L)及Glg(0、100、500、1000 ng/L)處理MIN6細胞。ELISA測定細胞內(nèi)cGMP含量,同時測定細胞上清液中Ins的含量。結(jié)果單純Glg處理組中,1000 ng/L組cGMP含量較500 ng/L組、100 ng/L組低(P0.05)。2.8 mmol/L Glu加不同濃度Glg處理組中,1000 ng/L Glg組cGMP含量較100 ng/L Glg組及0 ng/L Glg組低;500 ng/L Glg組較0 ng/L Glg組低(P0.05)。16.7 mmol/L Glu加不同濃度Glg處理組中,1000 ng/L Glg組cGMP含量較其他組低;500 ng/L Glg組較100 ng/L Glg組及0 ng/L Glg組低(P0.05)。單純Glg處理組中,隨Glg濃度升高,Ins分泌量升高(P0.05)。2.8 mmol/L Glu加不同濃度Glg處理組中,1000 ng/L Glg組Ins較100 ng/L Glg組及0 ng/L Glg組高;500 ng/L Glg組Ins較0 ng/L Glg組高(P0.05)。16.7 mmol/L Glu加不同濃度Glg處理組中,隨Glg濃度升高,Ins分泌量升高(P0.05)。結(jié)論 Glg可能通過濃度梯度的方式下調(diào)MIN6細胞內(nèi)cGMP的產(chǎn)生從而促進MIN6細胞Ins分泌,且這種作用有Glu依賴性。
[Abstract]:Objective to investigate the effects of different concentrations of glucagon (Glg) and glucose (Glu) on insulin (ins) secretion and intracellular cyclic guanosine (cGMP) signaling pathway in human islet 尾 cell line MIN6. Methods MIN6 cells were treated with different concentrations of Glu (0 2. 810. 7 mmol / L) and Glg (0 100 ~ 100 ~ 500 ~ 1 000 ng / L). The content of cGMP was measured by Elisa and the content of ins in supernatant of cells was also measured by enzyme-linked immunosorbent assay (Elisa). Results the cGMP content in 1000 ng / L group was lower than that in 100 ng / L group of 500 ng / L group (P0.05). 2.8 mmol / L Glu plus different concentrations of Glg group. The cGMP content in 1000 ng / L Glg group was lower than that in 100 ng / L Glg group and 0 ngL Glg group (P0.05). 16.7 mmol / L Glu content was significantly lower than that in 0 ng / L Glg group (P0.05). The content of cGMP in 1000ng / L Glg group was lower than that in other groups (P 0.05). The content of cGMP was lower in 500ng / L Glg group than that in 100ng / L Glg group and 0 ng / L Glg group (P0.05). The secretion of Ins increased with the increase of Glg concentration (P0.05) .2.8 mmol / L Glu + different concentration Glg treatment group. The ins of 1000 ng / L Glg group was higher than that of 100 ng / L Glg group and 0 ng / L Glg group, and the ins of 0 ng / L Glg group was higher than that of 0 ng / L Glg group (P0.05). 16.7 mmol / L Glu + different concentration Glg treatment group. The secretion of Ins increased with the increase of Glg concentration (P0.05). Conclusion Glg may down-regulate the production of cGMP in MIN6 cells and promote the secretion of Ins in MIN6 cells by concentration gradient, and this effect is Glu dependent.
