黃芩素、氨基胍干預(yù)下糖尿病大鼠類表皮樣生長因子結(jié)構(gòu)蛋白-7腦組織表達(dá)變化的研究

【摘要】:目的探討黃芩素、氨基胍干預(yù)對(duì)糖尿病大鼠腦組織中人類表皮樣生長因子結(jié)構(gòu)蛋白-7(EGFL-7)表達(dá)的影響。方法選取6~8周齡健康雄性SD大鼠40只,隨機(jī)分為正常對(duì)照(NC)組、糖尿病組、氨基胍組、黃芩素組,每組各10只。NC組予普通飲食喂養(yǎng);糖尿病組、氨基胍組、黃芩素組建立糖尿病大鼠模型,氨基胍組、黃芩素組分別予氨基胍、黃芩素溶液灌胃。檢測各組腦組織EGFL-7表達(dá)情況,觀察各組腦血管內(nèi)皮超微結(jié)構(gòu)。結(jié)果與NC組比較,糖尿病組FPG升高,胰島素降低(P0.05);與糖尿病組比較,氨基胍組和黃芩素組FPG降低,胰島素升高(P0.05)。NC組毛細(xì)血管腔規(guī)則,糖尿病組毛細(xì)血管腔不規(guī)則,部分狹窄變形,血管周圍可見大量水腫液,氨基胍組部分血管形態(tài)不規(guī)則,黃芩素組血管內(nèi)皮細(xì)胞結(jié)構(gòu)清晰;與NC組比較,糖尿病組EGFL-7表達(dá)陽性細(xì)胞數(shù)、相對(duì)灰度值升高[(0.41±0.18)%vs(19.20±2.56)%,(25.32±3.01)%vs(60.10±5.73)%,P0.05];與糖尿病組比較,氨基胍組和黃芩素組EGFL-7表達(dá)陽性細(xì)胞數(shù)、相對(duì)灰度值降低[(19.20±2.56)%vs(14.83±2.31)%、(13.89±3.08)%,(60.10±5.73)%vs(35.90±4.91)%、(34.12±5.04)%,P0.05]。結(jié)論黃芩素、氨基胍可影響糖尿病大鼠腦血管增生和重塑,其作用可能與降低腦組織EGFL-7表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effects of baicalin and aminoguanidine on the expression of human epidermal growth factor structural protein 7 (EGFL-7) in diabetic rats. Methods Forty healthy male SD rats aged 6 to 8 weeks were randomly divided into normal control (NC) group, diabetes group, aminoguanidine group, baicalein group, 10 rats in each group, and diabetes group, aminoguanidine group, baicalein group, diabetes group and aminoguanidine group respectively. The diabetic rats in baicalin group, aminoguanidine group and baicalin group were given aminoguanidine and baicalin solution respectively. The expression of EGFL-7 in brain tissue and the ultrastructure of cerebral vascular endothelium in each group were detected. Results compared with NC group, FPG increased and insulin decreased in diabetic group (P0.05), FPG in aminoguanidine group and baicalin group decreased, insulin increased (P0.05). Some stenosis and deformation, a large amount of edema around the vessels, irregular morphology of some vessels in aminoguanidine group, clear structure of vascular endothelial cells in baicalin group, and EGFL-7 positive cells in diabetic group compared with NC group, the number of EGFL-7 positive cells in diabetic group was higher than that in control group. The relative gray scale increased [(0.41 鹵0.18) vs (19.20 鹵2.56), (25.32 鹵3.01) vs (60.10 鹵5.73) P 0.05], compared with the diabetic group, the number of EGFL-7 positive cells in aminoguanidine group and baicalin group decreased [(19.20 鹵2.56) vs (14.83 鹵2.31), (13.89 鹵3.08), (60.10 鹵5.73) vs (35.90 鹵4.91), (34.12 鹵5.04)]. Conclusion baicalin and aminoguanidine can affect cerebral vascular hyperplasia and remodeling in diabetic rats, which may be related to the decrease of EGFL-7 expression in brain tissue.
