L型鈣離子通道阻滯劑貝尼地平對(duì)去卵巢小鼠骨代謝的研究

【摘要】:目的探討貝尼地平對(duì)去卵巢骨質(zhì)疏松模型小鼠的骨代謝影響。方法隨機(jī)選取鼠齡10周雌性小鼠30只,均分為假手術(shù)組,單純?nèi)ヂ殉步M,去卵巢加藥組。假手術(shù)組只去除卵巢周?chē)倭恐?單純?nèi)ヂ殉步M去除卵巢后灌胃等量生理鹽水,去卵巢加藥組去除卵巢后灌胃貝尼地平[20 mg/(kg·d),benidipine,BD]3個(gè)月。取小鼠股骨,經(jīng)4%多聚甲醛固定,脫鈣后分別進(jìn)行股骨下段石蠟切片伊紅蘇木素(HE)染色、轉(zhuǎn)錄因子RUNX2(runt-related transcription factor 2,Runx2)免疫組織化學(xué)染色及骨鈣素(osteocalcin,OCN)免疫熒光(immunofluorescence,IF)染色,并進(jìn)行顯微CT(μCT)掃描。結(jié)果股骨下段HE染色發(fā)現(xiàn)灌胃貝尼地平的去卵巢小鼠股骨下段骨小梁面積明顯高于單純?nèi)ヂ殉残∈蠼M(OVX組)(P0.05),差異具有統(tǒng)計(jì)學(xué)意義,和假手術(shù)組相比,去卵巢加藥組股骨下段骨小梁明顯降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);股骨下段RUNX2免疫組化染色發(fā)現(xiàn),和單純切卵巢組小鼠相比,貝尼地平治療組的蛋白表達(dá)量明顯上調(diào),差異具有統(tǒng)計(jì)學(xué)意義(P0.05),而各組中,假手術(shù)組RUNX2表達(dá)量仍然是最高的,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);通過(guò)骨鈣素免疫熒光染色發(fā)現(xiàn),貝尼地平治療組股骨下段骨鈣素表達(dá)量明顯高于去卵巢組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05),但是仍低于假手術(shù)組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);此外,通過(guò)股骨下段顯微CT掃描發(fā)現(xiàn),去卵巢小鼠灌胃貝尼地平組股骨下段骨密度(bone mineral density,BMD),骨小梁厚度(trabecular thickness,Tb.Th),骨小梁數(shù)量(trabecular number,Tb.N)均明顯高于去卵巢組,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05),但骨代謝指標(biāo)仍低于假手術(shù)組,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論貝尼地平能明顯減緩由于去卵巢引起的C57/BL6雌鼠骨量丟失,具有比較明顯的抗骨質(zhì)疏松作用。
[Abstract]:Objective to investigate the effect of benidipine on bone metabolism in ovariectomized osteoporosis mice. Methods Thirty female mice aged 10 weeks were randomly divided into sham-operation group, ovariectomized group and ovariectomized group. The sham operation group only removed a small amount of fat around the ovary, the simple ovariectomized group removed the ovary, and the ovariectomized group was given the same amount of normal saline after removing the ovary, and the ovariectomized plus drug group was given benidipine [20 mg/(kg] for 3 months. The femur of the mice was fixed with 4% paraformaldehyde, and then decalcified with eosylcyrrhizin (HE) staining, RUNX2(runt-related transcription factor _ 2 Runx _ 2) immunohistochemical staining and osteocalcin OCN-immunofluorescence (IFIFF) staining, and the microCTs were scanned by microCTs (渭 CTs). Results the area of trabecular bone in ovariectomized mice fed with benidipine was significantly higher than that in OVX group (P 0.05), and the difference was statistically significant compared with sham operation group. In the ovariectomized group, the trabecular bone of the lower femur was significantly decreased, the difference was statistically significant (P 0.05), and the expression of protein in benidipine treatment group was significantly higher than that in the simple ovariectomized group, and the RUNX2 immunohistochemical staining in the lower femur segment of the femur was significantly higher than that in the ovariectomized group. The difference was statistically significant (P 0.05), but the expression of RUNX2 in sham operation group was still the highest (P 0.05), which was found by osteocalcin immunofluorescence staining. The expression of osteocalcin in the lower femur of the benidipine group was significantly higher than that in the ovariectomized group, the difference was statistically significant (P 0.05), but still lower than that in the sham operation group (P 0.05). In ovariectomized mice, the bone mineral density, trabecular thickness, trabecular thickness and trabecular number Tb. N were significantly higher in the ovariectomized group than in the ovariectomized group. The difference was statistically significant (P 0.05), but the bone metabolism index was still lower than that in the sham operation group. The difference was statistically significant (P 0.05). Conclusion Benidipine can significantly reduce bone loss in ovariectomized C57/BL6 female rats and has obvious anti-osteoporosis effect.
