【摘要】：目的觀察左甲狀腺素鈉、兒茶素以及兩藥聯(lián)合治療對甲狀腺功能減退模型大鼠認(rèn)知能力的影響,對血清8-羥基脫氧鳥苷(8-OHd G)、對氧磷酶-1含量的影響以及海馬組織凋亡蛋白caspase-3含量的影響。方法選取50只成年SD雄性大鼠,隨機(jī)選取10只作為對照組,剩余40只用丙基硫氧密啶(PTU)誘導(dǎo)甲狀腺功能減退大鼠模型。PTU組、兒茶素組、左甲狀腺素鈉組(L-T4組)、左甲狀腺素鈉+兒茶素組(聯(lián)合用藥組)。采用Morris水迷宮系統(tǒng)進(jìn)行大鼠行為學(xué)檢測。采用酶聯(lián)免疫吸附試驗測定各組大鼠血清8-OHd G、對氧磷酶-1水平,采用Western blot測定各組大鼠海馬組織凋亡蛋白caspase-3的含量。結(jié)果一、行為學(xué)檢測與對照組比較,PTU組大鼠逃避潛伏期明顯延長,差異有統(tǒng)計學(xué)意義(P0.05);與PTU組比較,L-T4組、聯(lián)合用藥組大鼠逃避潛伏期均明顯縮短,差異有統(tǒng)計學(xué)意義(P0.05)。二、指標(biāo)檢測(1)與對照組比較,PTU組大鼠血清8-OHd G含量明顯升高,差異有統(tǒng)計學(xué)意義(P0.05);與PTU組比較,L-T4組、聯(lián)合用藥組大鼠血清8-OHd G含量明顯降低,差異有統(tǒng)計學(xué)意義(P0.05)。?與對照組比較,PTU組大鼠血清對氧磷酶-1含量明顯降低,差異有統(tǒng)計學(xué)意義(P0.05);與PTU組比較,L-T4組、聯(lián)合用藥組大鼠血清對氧磷酶-1含量明顯升高,差異有統(tǒng)計學(xué)意義(P0.05)。?與對照組比較,PTU組大鼠海馬組織凋亡蛋白caspase-3含量明顯升高,差異有統(tǒng)計學(xué)意義(P0.05);與PTU組比較,LT4組、聯(lián)合用藥組大鼠海馬組織凋亡蛋白caspase-3含量明顯降低,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論甲狀腺功能減退可降低機(jī)體的抗氧化應(yīng)激能力以及增加細(xì)胞的凋亡,左甲狀腺素鈉、兒茶素藥物治療可以改善氧化應(yīng)激、減少細(xì)胞的凋亡,改善大鼠的認(rèn)知能力,兩藥聯(lián)合使用具有協(xié)同的作用。
[Abstract]:Objective to observe the effects of levothyroxine sodium, catechin and combined therapy on the cognitive ability of hypothyroidism rats. The changes of 8-hydroxydeoxyguanosine guanosine 8-OHd (8-OHd), phosphatase-1 and apoptotic protein caspase-3 in hippocampal tissue were studied. Methods 50 adult Sprague-Dawley male rats were selected and 10 rats were randomly selected as control group. The remaining 40 rats were used to induce hypothyroidism model. PTU group and catechin group. The levothyroxine sodium group was treated with L-T4 and the levothyroxine sodium catechin group. The behavior of rats was detected by Morris water maze system. The levels of serum 8-OHd G and paraoxonase-1 were measured by enzyme-linked immunosorbent assay (Elisa), and the content of apoptotic protein caspase-3 (caspase-3) in hippocampal tissue of rats in each group was measured by Western blot. The results were as follows: (1) compared with the control group, the latency of escape in the PTU group was significantly longer than that in the control group, the difference was statistically significant (P 0.05), and that in the L-T4 group was significantly shorter than that in the PTU group (P 0.05). (2) compared with the control group, the serum 8-OHd G level in the control group was significantly higher than that in the control group (P 0.05), and that in the L-T4 group was significantly lower than that in the L-T4 group, and the difference was statistically significant (P 0.05). Compared with the control group, the level of serum paraoxonase-1 in the PTU group was significantly lower than that in the control group, the difference was statistically significant (P 0. 05), and that in the L-T4 group was significantly higher than that in the PTU group, and the difference was statistically significant (P 0. 05, P 0. 05, P 0. 05, P < 0. 05, P < 0. 05, P < 0. 05, P < 0. 05). Compared with the control group, the content of apoptotic protein caspase-3 in hippocampal tissue in PTU group was significantly higher than that in control group (P 0.05), and that in LT4 group was significantly lower than that in LT4 group (P 0.05). Conclusion hypothyroidism can decrease the ability of anti-oxidative stress and increase the apoptosis of cells. The treatment of levothyroxine sodium and catechin can improve the oxidative stress, decrease the apoptosis of cells, and improve the cognitive ability of rats. The combined use of the two drugs has synergistic effect.
