慢性錳中毒大鼠5-羥色胺、腦源性營養(yǎng)因子的表達變化

【摘要】:目的研究慢性錳中毒大鼠腦組織的5-羥色胺(5-HT)、腦源性營養(yǎng)因子(BDNF)的表達變化,了解慢性錳中毒大鼠抑郁行為及錳蓄積對腦組織5-HT、BDNF表達的影響。方法取80只SPF級SD雄性大鼠,分為2組:對照組和實驗組,實驗組又分為三個亞組:低錳組、中錳組、高錳組,分別腹腔注射Mncl2.4H2O 5 mg/kg、15 mg/kg、25 mg/kg,對照組予注射等量的生理鹽水,獲得慢性錳中毒大鼠模型。于第12周采用蔗糖水偏好試驗、敝箱實驗評估大鼠是否具有抑郁樣癥狀,電感耦合等離子體-原子發(fā)射光譜法(ICP-AES法)測定紋狀體錳含量。運用高效液相色譜法測定額葉、海馬5-HT含量,Western blot法檢測額葉、海馬的BDNF表達,并采用實時熒光定量PCR技術檢測BDNF、5-HT的限速酶色氨酸羥化酶2 (TPH2) mRNA的表達情況。結果1、蔗糖消耗試驗及敝箱實驗結果顯示,慢性錳中毒大鼠出現(xiàn)抑郁樣行為,高劑量組表現(xiàn)更加明顯,差別有統(tǒng)計學意義(均P0.05)。2、高效液相色譜法結果表明給錳組大鼠額葉及海馬BDNF表達隨錳濃度增加而下降明顯,其中高錳組表達最低,呈劑量依賴性,差別有統(tǒng)計學意義(均P0.05)。3. Western blot結果表明給錳組大鼠額葉及海馬BDNF表達隨錳濃度增加而下降明顯,其中高錳組表達最低,呈劑量依賴性,差別有統(tǒng)計學意義(均P0.05)。4、real-time PCR結果示錳暴露后大鼠額葉皮層及海馬的TPH2mRNA, BDNFmRNA表達水平均降低,且與錳中毒程度相關聯(lián),呈劑量依賴性,差別有統(tǒng)計學意義(均P0.05)。結論1、慢性錳中毒大鼠有抑郁樣行為存在;2、錳蓄積致額葉及海馬神經(jīng)元內(nèi)5-HT、BDNF表達下調(diào),慢性錳中毒大鼠伴發(fā)抑郁可能與腦組織5-HT、BDNF表達下調(diào)有關。
[Abstract]:Objective to study the chronic manganese poisoning rat brain serotonin 5- (5-HT), brain-derived neurotrophic factor (BDNF) expression, understanding chronic manganese poisoning Depression Behavior of rats and manganese accumulation of 5-HT on brain tissue, BDNF expression. Methods 80 SPF male SD rats were divided into 2 groups. The control group and the experimental group, the experimental group was divided into three subgroups: low manganese group, manganese group, high manganese group, intraperitoneal injection of Mncl2.4H2O 5 mg/kg, 15 mg/kg, 25 mg/kg, control group with normal saline injected, for chronic manganese poisoning rats. In twelfth weeks by sugarcane sucrose preference test, our experimental evaluation of rats with box depression like symptoms, inductively coupled plasma atomic emission spectrometry (ICP-AES) determination of manganese content in striatum. The use of HPLC method for the determination of 5-HT content in frontal lobe, hippocampus, frontal Western detection method of blot expression in hippocampus of BDNF, and a real Fluorescence quantitative PCR detection of BDNF, the rate limiting enzyme of tryptophan hydroxylase 2 (5-HT TPH2) expression of mRNA. Results 1, sucrose consumption test box and our experimental results show that the depression like behavior in rats of chronic manganese poisoning, the high dose group was more obvious, the difference was statistically significant (P0.05).2, the results showed that the HPLC method for manganese in rats and the expression of BDNF in hippocampus and frontal lobe decreased significantly with the increase of the concentration of manganese, high manganese group was the lowest in a dose-dependent manner, the difference was statistically significant (P0.05).3. Western blot analysis showed that Mn treated rats decreased the expression of BDNF in frontal lobe and hippocampus obviously when Mn concentration increased, the high manganese group was the lowest in a dose-dependent manner, the difference was statistically significant (P0.05).4, real-time PCR TPH2mRNA showed frontal cortex and hippocampus of rats exposed to manganese, the expression level of BDNFmRNA decreased, and Associated manganese poisoning, in a dose-dependent manner, the difference was statistically significant (P0.05). Conclusion: 1. Chronic manganese intoxication in rats with depression like behavior; 2, manganese accumulation induced by frontal and hippocampal neurons in 5-HT, BDNF expression, chronic manganese intoxication in rats with depression and brain tissue 5-HT. The down-regulation of BDNF.
