【摘要】：目的研究Toll樣受體2(TLR2)基因敲除對高脂飲食誘導(dǎo)的肥胖小鼠脂肪組織炎癥因子表達(dá)和細(xì)胞凋亡的影響。方法雄性C57BL6小鼠和TLR2基因敲除小鼠分為:正常對照組、肥胖組、TLR2基因敲除組和TLR2基因敲除肥胖組,并給予普通飲食或高脂飲食喂養(yǎng)。16周后檢測各組小鼠空腹血糖、甘油三酯、總膽固醇、低密度脂蛋白、高密度脂蛋白和游離脂肪酸水平。分光光度計(jì)法檢測小鼠脂肪組織Caspase3活性,Western blot檢測Bcl2、Bax、MyD88和p65NFκB蛋白相對表達(dá)量。熒光實(shí)時(shí)定量PCR檢測小鼠脂肪組織IL-6和TNF-αmRNA相對表達(dá)量。結(jié)果與正常對照組相比,肥胖組小鼠空腹血糖、甘油三酯、總膽固醇、低密度脂蛋白、高密度脂蛋白水平降低和游離脂肪酸水平升高;脂肪組織Caspase3活性、Bax蛋白、MyD88蛋白、p65NFκB蛋白、IL-6 mRNA和TNF-αmRNA相對表達(dá)量升高,Bcl2蛋白相對表達(dá)量減少。與肥胖組相比,基因敲除肥胖組小鼠空腹血糖、甘油三酯、總膽固醇、低密度脂蛋白、高密度脂蛋白水平降低和游離脂肪酸水平降低;脂肪組織Bcl2蛋白相對表達(dá)量增加,Caspase3活性、Bax蛋白、MyD88蛋白、p65NFκB蛋蛋白、IL-6 mRNA和TNF-αmRNA相對表達(dá)量減少。結(jié)論 TLR2基因敲除下調(diào)了高脂飲食誘導(dǎo)的肥胖小鼠脂肪組織炎癥因子的表達(dá)和細(xì)胞凋亡。
[Abstract]:Objective to study the effects of Toll like receptor 2 (TLR2) knockout on the expression of inflammatory factors and apoptosis in adipose tissue of obese mice induced by high fat diet.Methods male C57BL6 mice and TLR2 knockout mice were divided into three groups: normal control group, obese group and obese group. Fasting blood glucose and triglyceride were measured after feeding with normal diet or high-fat diet for .16 weeks.Total cholesterol, low density lipoprotein, high density lipoprotein and free fatty acid levels.The activity of Caspase3 in adipose tissue of mice was detected by spectrophotometer and the relative expression of Bcl2Bax-MyD88 and p65NF 魏 B protein was detected by Western blot.The relative expression of IL-6 and TNF- 偽 mRNA in adipose tissue of mice was detected by fluorescence real time quantitative PCR.Results compared with the control group, the levels of fasting blood glucose, triglyceride, total cholesterol, low density lipoprotein, high density lipoprotein and free fatty acid increased in obese group.The relative expression of IL-6 mRNA and TNF- 偽 mRNA in adipose tissue was increased and the relative expression of Bcl2 protein was decreased.Compared with the obese group, the fasting blood glucose, triglyceride, total cholesterol, low density lipoprotein, high density lipoprotein and free fatty acid were decreased in the gene knockout group.The relative expression of Bcl2 protein in adipose tissue increased the activity of Caspase3 and decreased the relative expression of IL-6 mRNA and TNF- 偽 mRNA in p65NF 魏 B egg protein.Conclusion TLR2 knockout can down-regulate the expression of inflammatory factors and apoptosis in adipose tissue of obese mice induced by high fat diet.
