控制性超促排卵周期多囊卵巢綜合征患者生長分化因子-9和骨形成蛋白-15的初步研究

【摘要】:目的:探討控制性超促排卵(COH)周期多囊卵巢綜合征(PCOS)患者卵巢顆粒細(xì)胞生長分化因子-9(GDF-9)、骨形成蛋白-15(BMP-15)的表達(dá)及其與體外受精-胚胎移植(IVF-ET)治療結(jié)局的關(guān)系。方法:選擇接受IVF-ET治療的PCOS患者45例,同期輸卵管因素不孕患者103例作為對照組。應(yīng)用免疫組織化學(xué)、Real-time PCR技術(shù)檢測患者卵巢顆粒細(xì)胞GDF-9、BMP-15蛋白和mRNA的表達(dá),并分析與IVF-ET的臨床結(jié)局關(guān)系。結(jié)果:GDF-9、BMP-15蛋白在PCOS組和對照組患者卵巢顆粒細(xì)胞胞質(zhì)均呈陽性表達(dá),PCOS患者顆粒細(xì)胞GDF-9 mRNA、BMP-15 mRNA的表達(dá)水平低于對照組,差異有統(tǒng)計學(xué)意義(P0.01)。多元線性回歸分析顯示,顆粒細(xì)胞GDF-9 mRNA、BMP-15 mRNA表達(dá)水平與優(yōu)質(zhì)胚胎數(shù)呈正相關(guān)關(guān)系(P0.05)。臨床妊娠組的顆粒細(xì)胞GDF-9mRNA、BMP-15 mRNA表達(dá)水平高于未妊娠組(P0.05)。結(jié)論:COH周期PCOS患者顆粒細(xì)胞GDF-9 mRNA、BMP-15 mRNA表達(dá)水平可能與胚胎發(fā)育潛能存在相關(guān)性。
[Abstract]:Objective: to investigate the expression of granulosa cell growth differentiation factor (GDF-9) and bone morphogenetic protein (BMP-15) in ovarian granulosa cell growth differentiation factor (GDF-9) and bone morphogenetic protein-15 (BMP-15) in patients with polycystic ovary syndrome (PCOSs) during the controlled ovarian hyperstimulation (COH) cycle and their relationship with the outcome of IVF-ETS treatment. There were 45 patients with PCOS who were treated with IVF-ET. The expression of GDF-9 BMP-15 and mRNA in ovarian granulosa cells was detected by immunohistochemical real-time PCR technique in 103 infertile patients with oviduct factor as control group. Results the expression level of GDF-9 mRNA-BMP-15 mRNA in granulosa cells of PCOS group and control group was lower than that in control group, and the expression of GDF-9 mRNA-BMP-15 protein in granulosa cells of PCOS group and control group was lower than that of control group. The difference was statistically significant (P 0.01). Multiple linear regression analysis showed that, The expression level of BMP-15 mRNA in granulosa cells was positively correlated with the number of high quality embryos. The expression level of BMP-15 mRNA in granulosa cells in clinical pregnancy group was higher than that in unpregnant group. Conclusion the expression level of GDF-9 mRNA-BMP-15 mRNA in granulosa cells of patients with PCOS in the cell cycle of 20 COH cycle may be related to the expression of BMP-15 mRNA in embryo embryos. Fetal development potential is related.
