【摘要】:目的探討中國漢族人群糖皮質(zhì)激素誘導(dǎo)轉(zhuǎn)錄因子1(GLCCI1)基因rs37973位點多態(tài)性與哮喘的相關(guān)性。方法應(yīng)用聚合酶鏈反應(yīng)(PCR)、DNA測序、哮喘控制測試評分(ACT)及肺功能測定的方法。結(jié)果不同種族人群GLCCI1基因rs37973位點等位基因頻率的比較差異有統(tǒng)計學(xué)意義(X~2=89.07,P0.05)。我國漢族哮喘人群及健康者rs37973位點均存在基因多態(tài)現(xiàn)象(表型包括AA、AG和GG三種基因型),各基因型分布頻率哮喘組與健康對照組比較差異有統(tǒng)計學(xué)意義(X~2=8.27,P0.05);A/G等位基因頻率比較差異有統(tǒng)計學(xué)意義(X~2=4.14,P0.05)。相對AA基因型而言,AG雜合型、GG+AG均能顯著增加哮喘發(fā)生的危險性。rs37973位點等位基因頻率與哮喘關(guān)系表明,相對A而言,突變G能顯著增加哮喘發(fā)生的危險性。在哮喘組3種基因型的ACT評分、第1秒用力呼氣容積(FEV_1)、第1秒用力呼氣容積占預(yù)計值的百分比(FEV_1/Pred%)、第1秒用力呼氣容積占用力肺活量的百分比(FEV_1/FVC%)比較差異無統(tǒng)計學(xué)意義(P均0.05)。結(jié)論我國漢族人群存在著GLCCI1基因rs37973位點多態(tài)性,且其多態(tài)性與哮喘患病相關(guān)。
[Abstract]:Objective to investigate the association of rs37973 polymorphism of glucocorticoid inducible transcription factor 1 (GLCCI1) gene with asthma in Chinese Han population. Results there were significant differences in allele frequencies of GLCCI1 rs37973 locus in different ethnic groups. There was a significant difference in the allele frequency of rs37973 locus of GLCCI1 gene. The rs37973 loci were found in both Chinese Han asthmatic population and healthy subjects. Because of polymorphism (phenotypes including AAAAG and GG genotypes), the frequency of genotype distribution in asthma group was significantly different from that in healthy control group. There was a significant difference in allele frequency of A- / G allele between the asthma group and the healthy control group. There was a statistically significant difference in the allele frequency of the A / G allele between the AA genotype and the AA genotype. The allelic frequency of allelic locus. Rs37973 and asthma were significantly increased by AG heterozygous GG AG, indicating that GG AG could significantly increase the risk of asthma. Compared with A, mutation G significantly increased the risk of asthma. In the asthma group, the ACT score of three genotypes, There was no significant difference in FEV volume at 1 seconds, FEV volume at 1 seconds as a percentage of predicted value, FEV 1 / Predg at 1 seconds and FEV1 / FVCC5.
