蛋白激酶C活性與糖尿病足截肢患者脛前動(dòng)脈鈣化積分的相關(guān)性

【摘要】:目的探討蛋白激酶C(PKC)活性與糖尿病足截肢患者脛前動(dòng)脈鈣化之間的相關(guān)性。方法選取2012年10月至2016年1月在江蘇大學(xué)附屬醫(yī)院骨科行糖尿病足大截肢住院治療的患者共60例,根據(jù)CT對(duì)脛前動(dòng)脈鈣化程度的檢測(cè)將患者分為輕度鈣化組(n=20)、中度鈣化組(n=20)及重度鈣化組(n=20)。通過(guò)患者臨床基線資料的采集以及脛前動(dòng)脈組織連續(xù)石蠟切片Von Kossa染色觀察鈣化演進(jìn),通過(guò)PepTag~汶Assay法檢測(cè)組織細(xì)胞膜PKC活性,通過(guò)Western blot檢測(cè)對(duì)PKCα亞型進(jìn)行半定量分析。結(jié)果糖尿病足截肢患者脛前動(dòng)脈Von Kossa染色與相應(yīng)的動(dòng)脈CT掃描結(jié)果一致:脛前動(dòng)脈斑塊內(nèi)的鈣鹽沉積呈點(diǎn)灶狀分布,而且隨著脛前動(dòng)脈內(nèi)斑塊的不斷擴(kuò)大乃至管腔閉塞,鈣顆粒的密度也有不同程度的增加,但始終呈點(diǎn)灶狀分布的微鈣化。定量檢測(cè)及Pearson相關(guān)性分析顯示PKC活性與糖尿病足脛前動(dòng)脈鈣化積分正相關(guān)(r=0.931,P0.001),進(jìn)一步分析顯示可能是PKCα亞型發(fā)揮了促進(jìn)鈣化演進(jìn)的作用。結(jié)論 PKCα亞型的活化可能與糖尿病動(dòng)脈斑塊內(nèi)鈣化的演進(jìn)存在正相關(guān),其水平可能對(duì)針對(duì)性干預(yù)血管鈣化有著一定的價(jià)值。
[Abstract]:Objective to investigate the correlation between protein kinase C kinase (PKC) activity and calcification of anterior tibial artery in diabetic foot amputation patients. Methods 60 patients with diabetic foot amputation were treated with diabetic foot amputation from October 2012 to January 2016 in Department of Orthopaedics, affiliated Hospital of Jiangsu University. The patients were divided into mild calcified group, moderate calcified group and severe calcified group according to CT examination of the calcification degree of anterior tibial artery. The clinical baseline data were collected and the Von Kossa staining of the tibial anterior artery tissue was used to observe the progress of calcification. The PKC activity of tissue membrane was measured by PepTag-Wen Assay assay. Results the results of Von Kossa staining of anterior tibial artery in diabetic foot amputation were consistent with those of CT scan. The calcium deposition in the plaque of anterior tibial artery was focal distribution. Moreover, with the enlargement of plaque in anterior tibial artery and the occlusion of lumen, the density of calcium granules increased in varying degrees. Quantitative analysis and Pearson correlation analysis showed that PKC activity was positively correlated with calcification integral of anterior tibial artery of diabetic foot. Further analysis showed that PKC 偽 subtype played a role in promoting calcification progression. Conclusion the activation of PKC 偽 subtype may be positively related to the progression of calcification in diabetic artery plaque. Its level may have certain value for intervention of vascular calcification.
