脂肪酸參與糖尿病狀態(tài)下肝細(xì)胞上CYP1A2功能與表達(dá)的上調(diào)

【摘要】:目的糖尿病上調(diào)肝臟CYP1A2的功能與表達(dá),但機(jī)制尚未明確。本研究擬從脂肪酸角度,通過(guò)體外研究初步探索糖尿病上調(diào)肝臟CYP1A2功能與表達(dá)的作用機(jī)制。方法通過(guò)測(cè)定CYP1A2底物非那西丁代謝水平考察肝細(xì)胞HepG2和Fa2N-4細(xì)胞上CYP1A2的酶活性,通過(guò)熒光實(shí)時(shí)定量PCR方法檢測(cè)細(xì)胞上CYP1A2 mRNA表達(dá)水平。分別用糖尿病大鼠(Ⅰ型和Ⅱ型)和正常大鼠血清培養(yǎng)HepG2細(xì)胞48 h,考察細(xì)胞上CYP1A2的功能。在培養(yǎng)基中加入不同濃度的飽和脂肪酸(軟脂酸和硬脂酸)和不飽和脂肪酸(油酸和亞油酸)培養(yǎng)HepG2和Fa2N-4細(xì)胞48 h,測(cè)定細(xì)胞上CYP1A2的功能與表達(dá)。結(jié)果Ⅰ型和Ⅱ型糖尿病大鼠血清培養(yǎng)的HepG2細(xì)胞上CYP1A2酶活性明顯增高。高濃度的脂肪酸均可增加2種細(xì)胞上CYP1A2功能與表達(dá)。結(jié)論糖尿病狀態(tài)下脂肪酸濃度的異常變化可能是影響肝臟CYP1A2功能與表達(dá)上調(diào)的原因之一,本研究為進(jìn)一步探索糖尿病狀態(tài)下肝臟CYP1A2改變機(jī)制提供了一定的基礎(chǔ)。
