【摘要】：目的研究絲膠對2型糖尿病大鼠肝臟胰島素受體底物(IRS)-2表達的影響。方法 36只雄性SD大鼠隨機分為正常對照組、糖尿病模型組、絲膠治療組,每組12只。采用鏈脲佐菌素連續(xù)腹腔注射法制作2型糖尿病大鼠模型。待模型成功建立后,絲膠治療組大鼠給予絲膠(2.4 g·kg~(-1)·d~(-1))灌胃。SP免疫組織化學染色、免疫印跡法和RT-PCR檢測大鼠肝臟組織IRS-2的表達。結果絲膠可明顯提高糖尿病模型大鼠肝臟組織IRS-2陽性細胞數(shù)、IRS-2蛋白和mRNA的表達(P0.01)。結論絲膠可通過上調(diào)糖尿病大鼠肝臟IRS-2的表達,影響胰島素的信號轉導和改善胰島素抵抗,起到降低血糖的作用。
[Abstract]:Objective to study the effect of sericin on the expression of insulin receptor substrate IRS-2 in liver of type 2 diabetic rats. Methods 36 male SD rats were randomly divided into normal control group, diabetic model group and sericin treatment group. The model of type 2 diabetic rats was established by continuous intraperitoneal injection of streptozotocin. After the successful establishment of the model, the rats in the sericin treatment group were given sericin 2.4 g 路kg ~ (-1) 路d ~ (-1)) by intragastric perfusion with immunohistochemical staining of SP. Results sericin could significantly increase the number of IRS-2 positive cells and the expression of IRS-2 protein and mRNA in the liver of diabetic rats. Conclusion sericin can up-regulate the expression of IRS-2 in the liver of diabetic rats. Affect insulin signal transduction and improve insulin resistance, play a role in reducing blood sugar.
