運(yùn)動(dòng)與白藜蘆醇對(duì)老年肥胖大鼠視黃醇結(jié)合蛋白4、血糖和胰島素敏感性的影響

【摘要】:目的探討運(yùn)動(dòng)與白藜蘆醇對(duì)老年肥胖大鼠內(nèi)臟脂肪組織視黃醇結(jié)合蛋白4(RBP4)mRNA和蛋白表達(dá)、血漿RBP4濃度、血糖及胰島素敏感性的影響。方法建立SD老年肥胖大鼠模型,選取自然生長(zhǎng)老年大鼠6只為正常組,將24只肥胖鼠隨機(jī)分為肥胖對(duì)照、運(yùn)動(dòng)、白藜蘆醇、運(yùn)動(dòng)+白藜蘆醇4組。白藜蘆醇為52.5 mg/(kg·d)灌胃,5次/周,持續(xù)干預(yù)8周。干預(yù)后測(cè)血糖、胰島素敏感性、內(nèi)臟脂肪組織RBP4 mRNA表達(dá)(qRT-PCR)、蛋白表達(dá)(Western blot)及血漿RBP4濃度(ELISA)。結(jié)果 8周干預(yù)后,(1)RBP4 mRNA表達(dá):肥胖組(2.63±0.45)高于正常組(2.10±0.15)(P0.05);白藜蘆醇組(1.84±0.33)、運(yùn)動(dòng)組(1.91±0.15)和運(yùn)動(dòng)+白藜蘆醇組(2.13±0.11)低于肥胖組(P0.05,P0.01)。(2)RBP4蛋白表達(dá):肥胖組(1.346±0.025)高于正常組(1.196±0.017)(P0.01);運(yùn)動(dòng)組(1.025±0.006)低于肥胖組(P0.01)、白藜蘆醇組(0.735±0.015)和運(yùn)動(dòng)+白藜蘆醇組(0.701±0.018)低于運(yùn)動(dòng)組(P0.01);運(yùn)動(dòng)+白藜蘆醇組低于白藜蘆醇組(P0.05)。(3)血漿RBP4濃度:肥胖組[(16.00±1.54)μg/L]高于正常組[(13.02±2.20)μg/L](P0.01);運(yùn)動(dòng)組[(14.76±1.56)μg/L]低于肥胖組,但差異無(wú)統(tǒng)計(jì)學(xué)意義;白藜蘆醇組[(13.59±0.07)μg/L]和運(yùn)動(dòng)+白藜蘆醇組[(12.98±1.69)μg/L]低于肥胖組(P0.05,P0.01)。(4)血糖:肥胖組[(17.93±6.09)mmol/L]高于正常組[(11.64±3.57)mmol/L](P0.01);運(yùn)動(dòng)組[(13.36±1.82)mmol/L]低于肥胖組(P0.05),白藜蘆醇組[(15.24±2.19)mmol/L]、運(yùn)動(dòng)+白藜蘆醇組[(13.95±2.26)mmol/L]低于肥胖組,但差異無(wú)統(tǒng)計(jì)學(xué)意義。(5)胰島素敏感性:肥胖組(0.37±0.02)低于正常組(0.39±0.02)(P0.05);白藜蘆醇組(0.38±0.01)、運(yùn)動(dòng)組(0.38±0.02)高于肥胖組,但差異無(wú)統(tǒng)計(jì)學(xué)意義,運(yùn)動(dòng)+白藜蘆醇組(0.39±0.15)高于肥胖組(P0.05)。上述指標(biāo)(除蛋白表達(dá)外)干預(yù)各組間差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論老年肥胖大鼠內(nèi)臟脂肪組織RBP4 mRNA、蛋白表達(dá)、血漿RBP4濃度及血糖均增高,胰島素敏感性降低;運(yùn)動(dòng)與白藜蘆醇均能降低其RBP4 mRNA、蛋白表達(dá)和血漿濃度;單純運(yùn)動(dòng)能明顯降低其血糖,運(yùn)動(dòng)結(jié)合白藜蘆醇能明顯提高其胰島素敏感性。在降低蛋白表達(dá)、血漿RBP4濃度和提高胰島素敏感性中,運(yùn)動(dòng)結(jié)合白藜蘆醇表現(xiàn)出協(xié)同性。
[Abstract]:Objective to investigate the effects of exercise and resveratrol on the expression of retinol binding protein 4 (RBP4N) mRNA and protein, plasma RBP4 concentration, blood glucose and insulin sensitivity in visceral adipose tissue of aged obese rats. Six aged naturally growing rats were selected as normal group. 24 obese rats were randomly divided into 4 groups: obese control group, exercise group, resveratrol group and exercise group. Resveratrol was given intragastrically for 5 times a week. Continuous intervention for 8 weeks. Blood glucose and insulin sensitivity were measured after intervention. Results the expression of RBP4 mRNA in visceral adipose tissue was significantly higher than that in normal group (2.63 鹵0.45), resveratrol group was 1.84 鹵0.33, exercise group was 1.91 鹵0.15) and exercise group was 2.13 鹵0.11) after 8 weeks of intervention, the expression of RBP4 mRNA was significantly lower in obese group than in normal group (2.63 鹵0.45), while in resveratrol group (1.84 鹵0.33), exercise group (1.91 鹵0.15) and exercise group (2.13 鹵0.11) were significantly lower than those in normal group. The expression of RBP4 protein in obese group (1.346 鹵0.025) was higher than that in normal group (1.196 鹵0.017), in exercise group (1.025 鹵0.006) was lower than that in obese group (0.735 鹵0.015) and in exercise group (0.701 鹵0.018) was lower than that in exercise group. Degree: the obese group [16.00 鹵1.54 渭 g / L] was higher than the normal group [13.02 鹵2.20 渭 g / L] P0.01g / L; the exercise group [14.76 鹵1.56 渭 g / L] was lower than the obese group. But there was no significant difference in blood glucose: the blood glucose levels in the resveratrol group [13.59 鹵0.07 渭 g / L] and the exercise resveratrol group [12.98 鹵1.69 渭 g / L] were lower than those in the obese group (P 0.05 P 0.01g / L): the obese group [17.93 鹵6.09 mmol / L] was higher than the normal group [11.64 鹵3.57 mmol / L] P0.01 / L; the exercise group [13.36 鹵1.82 mmol / L] was lower than the obese group; the resveratrol group [15.24 鹵2.19 mmol / L], the exercise group [13.95 鹵2.26 mmol / L] were lower than the obese group. But the insulin sensitivity of obese group was 0.37 鹵0.02) lower than that of normal group 0.39 鹵0.02P0.05, resveratrol group 0.38 鹵0.01g, exercise group 0.38 鹵0.02), but the difference was not statistically significant. Exercise resveratrol group (0.39 鹵0.15) was higher than obese group (P 0.05). There was no significant difference between the above indexes (except protein expression). Conclusion the expression of RBP4 mRNAs, protein expression, plasma RBP4 concentration and blood sugar in visceral adipose tissue of aged obese rats were increased. Both exercise and resveratrol could decrease RBP4 mRNAs, protein expression and plasma concentration, exercise alone could significantly decrease blood glucose, exercise combined with resveratrol could significantly increase insulin sensitivity. Exercise combined with resveratrol showed synergism in plasma RBP4 concentration and insulin sensitivity.
