【摘要】：目的:探討前列腺癌(prostate cancer,PCa)患者外周血淋巴細(xì)胞(peripheral blood lymphocyte,PBL)中perforin和granzyme-B的表達(dá)水平及意義。方法:運用熒光定量RT-PCR檢測60例PCa患者PBL中perforin和granzyme-B的m RNA的表達(dá)水平,同期檢測40例良性前列腺增生(benign prostatic hyperplasia,BPH)患者及20例健康者作為對照。結(jié)果:PCa組PBL中perforin和granzyme-B m RNA的表達(dá)水平較BPH組及健康對照組均明顯下降(P0.05);高病理分級組表達(dá)水平均低于低病理分級組(P0.05);高臨床分期組表達(dá)水平低于低臨床分期組(P0.05)。結(jié)論:PCa患者PBL中perforin和granzyme-B m RNA表達(dá)水平較對照組下降,且與病理分級和臨床分期負(fù)相關(guān),提示PCa的發(fā)生發(fā)展與患者免疫狀態(tài)下降有關(guān)。
[Abstract]:Objective: to investigate the expression and significance of perforin and granzyme-B in peripheral blood lymphocytes (peripheral blood lymphocyte,PBL) of patients with prostate cancer (prostate cancer,PCa). Methods: the expression of m RNA of perforin and granzyme-B in PBL of 60 patients with PCa was detected by fluorescence quantitative RT-PCR, and 40 patients with benign prostatic hyperplasia (benign prostatic hyperplasia,BPH) and 20 healthy controls were detected at the same time. Results: the expression levels of perforin and granzyme-B m RNA in PBL of PCa group were significantly lower than those of BPH group and healthy control group (P 0.05), the expression levels of perforin and granzyme-B m RNA in high pathological grade group were lower than those in low pathological grade group (P 0.05), and those in high clinical stage group were lower than those in low clinical stage group (P 0.05). Conclusion: the expression of perforin and granzyme-B m RNA in PBL of patients with PCa is lower than that of the control group, and is negatively correlated with pathological grade and clinical stage, suggesting that the occurrence and development of PCa is related to the decrease of immune status.
