【摘要】：目的探討膀胱尿路上皮癌(BUCC)中第10號染色體同源丟失性磷酸酶-張力蛋白基因(PTEN)表達與其基因啟動子甲基化的關(guān)系。方法應(yīng)用Western blot、甲基化特異性聚合酶鏈反應(yīng)(MSP),檢測41例BUCC和18例正常膀胱組織中PTEN蛋白的表達水平及其基因啟動子甲基化狀態(tài),并分析兩者的相關(guān)性。結(jié)果BUCC中PTEN蛋白表達低于正常膀胱組織(P0.05),而BUCC中PTEN基因啟動子甲基化率為53.66%(22/41),高于正常膀胱組織的0.00%(0/18)(P0.05);與甲基化陰性組的BUCC組織相比,22例甲基化陽性組的PTEN表達水平降低(P0.05)。結(jié)論 BUCC中PTEN蛋白的表達水平下調(diào),這與其基因啟動子高甲基化密切相關(guān)。
[Abstract]:Objective to investigate the relationship between the expression of homologous lost phosphatase-tensin gene (PTEN) on chromosome 10 and the methylation of its gene promoter in bladder and urinary tract epithelial carcinoma (BUCC). Methods the expression of PTEN protein and its promoter methylation status in 41 cases of BUCC and 18 cases of normal bladder tissue were detected by Western blot, methylated specific polymerase chain reaction (MSP), and the correlation between them was analyzed.
