小鼠頸部迷走神經(jīng)放電采集方法在腎缺血再灌注實(shí)驗(yàn)中的運(yùn)用

【摘要】:目的觀察研究C57BL/6J小鼠腎缺血再灌注期間頸部迷走神經(jīng)放電變化,并介紹其迷走神經(jīng)放電采集方法與技巧。方法雄性C57BL/6J小鼠8只,8~10周齡,體質(zhì)量22~26 g,沿頸正中線切開皮膚,逐層分離至頸迷走神經(jīng),用鉑金電極頭引導(dǎo)神經(jīng)放電并記錄其放電頻率;繼而進(jìn)行腹部手術(shù),通過(guò)夾閉雙側(cè)腎門缺血25 min后再恢復(fù)血流30 min,該過(guò)程持續(xù)記錄其迷走神經(jīng)放電頻率變化。結(jié)果與基礎(chǔ)放電頻率相比,腎缺血期間迷走神經(jīng)放電頻率由(520±52)Hz上升至(632±59)Hz(n=8,P0.05),再灌注后15 min內(nèi)放電頻率進(jìn)一步升高至(672±58)Hz(n=8,P0.05),之后逐步下降至(544±51)Hz(n=8,P0.05)。結(jié)論腎缺血再灌注期間小鼠頸部迷走神經(jīng)放電頻率較基礎(chǔ)放電頻率明顯上升,其原因可能涉及缺血再灌注時(shí)自身的神經(jīng)保護(hù)性機(jī)制。
[Abstract]:Objective to observe and study the changes of cervical vagus nerve discharge during renal ischemia-reperfusion in C57BL/6J mice, and to introduce the methods and techniques of vagus nerve discharge collection. Methods eight male C57BL/6J mice, 8 脳 10 weeks old and weighing 22 脳 26 g, were cut off along the median cervical line and separated into cervical vagus nerve layer by layer. The discharge frequency of vagus nerve was guided by platinum electrode head and recorded, and then abdominal surgery was performed to record the changes of vagus nerve discharge frequency by clamping bilateral hilar ischemia for 25 min and then resuming blood flow for 30 min,.
