著色性干皮病C組基因蛋白缺失對膀胱癌細胞自噬的影響

【摘要】:目的探討膀胱癌細胞中著色性干皮病C組基因(xeroderma pigmentosum group C,XPC)蛋白缺失對自噬的影響。方法利用shRNA策略,建立穩(wěn)定干擾XPC的膀胱癌T24細胞模型,蛋白印跡技術(shù)檢測干擾XPC后自噬表征蛋白LC3Ⅱ的表達,瞬時轉(zhuǎn)染GFP-LC3,熒光顯微鏡觀察細胞熒光聚集情況,CCK-8法檢測細胞的增殖狀況,免疫印跡技術(shù)檢測自噬及凋亡相關(guān)蛋白的表達。結(jié)果成功建立干擾XPC的膀胱癌T24細胞模型;干擾XPC后,膀胱癌T24細胞自噬小體的形成[(1.53±0.81)%vs(3.30±0.70)%,P0.05]明顯減少,順鉑作用下膀胱癌T24細胞自噬小體的形成[(2.19±0.37)%vs(3.20±0.29)%,P0.05]明顯減少;順鉑促進XPC缺失后的膀胱癌T24細胞自噬;XPC缺失后的自噬為非保護性自噬。結(jié)論 XPC蛋白參與調(diào)控膀胱癌T24細胞的自噬。
