【摘要】：目的研究FK506結(jié)合蛋白51(FK506Binding Protein51, FKBP51)在膀胱尿路上皮癌組織中的表達(dá)情況,并分析其與膀胱尿路上皮癌臨床資料及病理特征之間的關(guān)系,以探索FKBP51在膀胱尿路上皮癌發(fā)生發(fā)展中的作用。 方法隨機(jī)選取中南大學(xué)湘雅二醫(yī)院病理科存檔的膀胱癌蠟塊標(biāo)本中2011年1月至2013年12月期間手術(shù)方式為根治性膀胱術(shù)且病理證實(shí)為尿路上皮癌的標(biāo)本65例,所有患者術(shù)前均未行化療或放療。其中男性49例,女性16例,年齡范圍46.0-80.0歲,平均年齡64.2±8.6歲,中位年齡為66歲,年齡60歲：39例,60歲：26例。根據(jù)腫瘤大小：直徑≤3cm：23例,直徑3cm：42例。按2004年WHO組織學(xué)分級(jí)：低分級(jí)(高分化)乳頭狀尿路上皮癌41例,高分級(jí)(低分化)乳頭狀尿路上皮癌24例。按國際抗癌聯(lián)盟2009年第7版TNM分期對(duì)組織標(biāo)本進(jìn)行分期,其中非肌層浸潤性膀胱尿路上皮癌(包括Tis、Ta及T1期)18例,肌層浸潤性膀胱尿路上皮癌(包括T2及其以上)47例。按淋巴結(jié)是否轉(zhuǎn)移劃分,其中轉(zhuǎn)移16例,未轉(zhuǎn)移49例。同時(shí)收集15例正常膀胱組織,為對(duì)照組。應(yīng)用免疫組化技術(shù)檢測FKBP51蛋白在所研究標(biāo)本組織中的表達(dá)情況,并通過統(tǒng)計(jì)學(xué)方法分析其表達(dá)水平與膀胱尿路上皮癌臨床資料及病理特征的關(guān)系。 結(jié)果FKBP51蛋白在所有正常膀胱組織及癌組織中均表達(dá),但膀胱路上皮癌組織中表達(dá)比正常組織中表達(dá)明顯增高(p0.01),其表達(dá)水平與性別、年齡及腫瘤大小無關(guān)(p0.05),與腫瘤臨床分期(p0.05)、病理分級(jí)(p0.05)及淋巴結(jié)轉(zhuǎn)移(p0.05)相關(guān)。 結(jié)論1FKBP51蛋白在正常膀胱組織中低表達(dá),在膀胱尿路上皮癌組織中呈高表達(dá),這種差異性表達(dá)提示FKBP51可能與膀胱尿路上皮癌的發(fā)生相關(guān)。 2.FKBP51蛋白在膀胱尿路上皮癌組織中表達(dá)水平與其病理分級(jí)和臨床分期相關(guān),提示FKBP51蛋白可作為評(píng)估膀胱尿路上皮癌惡性程度的生物學(xué)標(biāo)志物,但具體特異性及敏感性有待進(jìn)一步研究。
[Abstract]:Objective to study the expression of FK506 binding protein 51 (FK506Binding Protein51, FKBP51) in bladder urothelial carcinoma and its relationship with clinical data and pathological features. To explore the role of FKBP51 in the occurrence and development of bladder urothelial carcinoma. Methods from January 2011 to December 2013, 65 cases of bladder cancer were randomly selected from the paraffin specimens of Department of Pathology of Xiangya second Hospital of Central South University as radical cystectomy and pathologically proved urothelial carcinoma. All patients were not treated with chemotherapy or radiotherapy before operation. There were 49 males and 16 females with an age range of 46.0-80.0 years, with an average age of 64.2 鹵8.6 years.
