【摘要】：目的： 觀察EMT相關(guān)表型分子E-cadherin、N-cadherin蛋白表達(dá)在腎透明細(xì)胞癌中的變化，探討EMT與腎透明細(xì)胞癌分期、分級(jí)之間的相關(guān)性。為臨床進(jìn)一步了解腎癌的發(fā)生發(fā)展及為判斷腎癌預(yù)后的研究提供理論依據(jù)。 方法： 收集腎透明細(xì)胞癌組織46例和癌旁正常組織10例制作組織芯片，應(yīng)用免疫組織化學(xué)鏈霉菌抗生物素蛋白-過(guò)氧化物酶連結(jié)(sP)法檢測(cè)E-cadherin、N-cadherin蛋白表達(dá)。 結(jié)果： 腎透明細(xì)胞癌E-cadherin陽(yáng)性表達(dá)明顯降低，同時(shí)N-cadherin陽(yáng)性表達(dá)明顯升高，E-cadherin與N-cadherin表達(dá)呈顯著相關(guān)性(P0.05)； E-cadherin的陽(yáng)性表達(dá)率與腎透明細(xì)胞癌的臨床分期、組織分化及有無(wú)淋巴結(jié)轉(zhuǎn)移有關(guān)（P＜0.05）。而N-cadherin的陽(yáng)性表達(dá)率與腎透明細(xì)胞癌的臨床分期、組織分化無(wú)明顯相關(guān)性（P＞0.05），，但與有無(wú)淋巴結(jié)轉(zhuǎn)移有相關(guān)性（P＜0.01）。 結(jié)論： 1、ccRCC發(fā)生發(fā)展過(guò)程中存在EMT，EMT參與ccRCC形成過(guò)程，并與腎透明細(xì)胞癌惡性程度有相關(guān)性。 2、N-cadherin的高表達(dá)和E-cadherin的低表達(dá)與ccRCC的發(fā)生發(fā)展，淋巴轉(zhuǎn)移有關(guān)。 3、N-cadherin的表達(dá)與ccRCC的預(yù)后有關(guān)
[Abstract]:Aim: to observe the expression of E-cadherin protein in renal clear cell carcinoma (RCC) and to investigate the correlation between EMT and stage and grade of renal clear cell carcinoma (RCC). To further understand the occurrence and development of renal cell carcinoma and to predict the prognosis of renal cell carcinoma. Methods: tissue microarray was made in 46 cases of renal clear cell carcinoma and 10 cases of adjacent normal tissues. The expression of E-cadherin protein was detected by immunohistochemical streptavidin-biotin peroxidase (sP) method. Results: the positive expression of E-cadherin was significantly decreased and the expression of N-cadherin was significantly increased in renal clear cell carcinoma. The expression of E-cadherin and N-cadherin was significantly correlated (P0.05). The positive expression rate of E-cadherin was related to the clinical stage, tissue differentiation and lymph node metastasis of renal clear cell carcinoma (P < 0. 05). However, the positive expression rate of N-cadherin was not correlated with clinical stage and histological differentiation of renal clear cell carcinoma (P > 0. 05), but correlated with lymph node metastasis (P < 0. 01). Conclusion: 1 EMT,EMT is involved in the formation of ccRCC and is related to the malignancy of renal clear cell carcinoma. 2the high expression of N-cadherin and the low expression of E-cadherin are related to the occurrence and development of ccRCC and lymphatic metastasis. 3The expression of N-cadherin is related to the prognosis of ccRCC.
