【摘要】：目的:探討正常前列腺組織不同區(qū)帶來(lái)源的前列腺間質(zhì)細(xì)胞對(duì)前列腺癌細(xì)胞生長(zhǎng)的影響及其作用機(jī)制。方法:以激素非依懶性前列腺癌細(xì)胞系DU145細(xì)胞為研究對(duì)象,取新鮮的正常前列腺外周帶(PZ)和移行帶(TZ)組織,提取間質(zhì)細(xì)胞并體外培養(yǎng)。收集不同區(qū)帶來(lái)源的間質(zhì)細(xì)胞培養(yǎng)上清作為條件培養(yǎng)液培養(yǎng)DU145細(xì)胞,CCK8法測(cè)定腫瘤細(xì)胞的生長(zhǎng)曲線,臺(tái)盼藍(lán)染色測(cè)定細(xì)胞數(shù)量及活力,細(xì)胞劃痕實(shí)驗(yàn)測(cè)定細(xì)胞侵襲性,Western印跡法測(cè)定間質(zhì)細(xì)胞對(duì)腫瘤細(xì)胞糖代謝關(guān)鍵酶的影響。結(jié)果:1PZ間質(zhì)細(xì)胞的條件培養(yǎng)液能促進(jìn)腫瘤細(xì)胞的生長(zhǎng),而TZ間質(zhì)細(xì)胞的條件培養(yǎng)液抑制腫瘤細(xì)胞的生長(zhǎng);2PZ間質(zhì)細(xì)胞的條件培養(yǎng)液明顯增加腫瘤細(xì)胞糖代謝關(guān)鍵酶己糖激酶2(HK-2)、丙酮酸激酶2(PKM-2)、乳酸脫氫酶(LDHA)、丙酮酸脫氫酶(PDH)的表達(dá),而TZ則抑制上述酶的表達(dá)。結(jié)論:前列腺不同來(lái)源的間質(zhì)細(xì)胞對(duì)腫瘤細(xì)胞的生長(zhǎng)影響不同,其機(jī)制可能與不同來(lái)源的間質(zhì)細(xì)胞對(duì)糖酵解代謝的影響不同有關(guān)。
[Abstract]:Aim: to investigate the effects of prostatic stromal cells from different zones of normal prostate tissue on the growth of prostate cancer cells and its mechanism. Methods: the stromal cells of DU145 cell line were isolated and cultured in vitro. The fresh (PZ) and (TZ) tissues of the normal prostate were collected and cultured in vitro. Mesenchymal cell culture supernatants from different zones were collected as conditioned medium to culture DU145 cells. The growth curves of tumor cells were measured by CCK8 assay, the number and viability of tumor cells were determined by trypan blue staining, and the invasiveness of tumor cells was measured by cell scratch assay. The effects of interstitial cells on the key enzymes of glucose metabolism in tumor cells were determined by Western blot. Results: the conditioned medium of 1PZ stromal cells could promote the growth of tumor cells, while the conditioned medium of TZ stromal cells inhibited the growth of tumor cells. The expression of hexokinase 2 (HK-2), pyruvate kinase 2 (PKM-2) and lactate dehydrogenase (LDHA),) pyruvate dehydrogenase (PDH) were significantly increased in the conditioned medium of 2PZ mesenchymal cells. TZ inhibited the expression of these enzymes. Conclusion: the effects of stromal cells from different prostatic sources on the growth of tumor cells may be related to the different effects of stromal cells from different prostatic sources on glycolytic metabolism.
