熒光差異雙向凝膠電泳篩選腎透明細(xì)胞癌及癌旁組織中的差異表達(dá)蛋白

【摘要】:目的尋找腎癌相關(guān)腫瘤標(biāo)記物,用于腎癌臨床診斷、藥物治療靶點(diǎn)的選擇及預(yù)后監(jiān)測(cè)。方法采用熒光差異雙向凝膠電泳技術(shù)篩選15例腎透明細(xì)胞癌及癌旁正常腎組織差異表達(dá)蛋白質(zhì)點(diǎn)。統(tǒng)計(jì)學(xué)采用t檢驗(yàn)進(jìn)行定量比較,以兩組蛋白含量比值在1.5倍及以上,P0.05,被認(rèn)為是差異表達(dá)蛋白質(zhì)點(diǎn)。用基質(zhì)輔助激光解吸電離飛行時(shí)間質(zhì)譜或串聯(lián)質(zhì)譜技術(shù)進(jìn)行差異表達(dá)蛋白質(zhì)點(diǎn)鑒定。結(jié)果共篩選分離出27個(gè)差異表達(dá)蛋白質(zhì)點(diǎn),經(jīng)質(zhì)譜技術(shù)共成功鑒定了26種蛋白質(zhì),11種蛋白質(zhì)在腎癌組織中上調(diào),15種蛋白質(zhì)下調(diào)。其功能覆蓋細(xì)胞活動(dòng)的多個(gè)方面,其中線粒體短/支鏈特異性;o酶A脫氫酶、醛糖1-表異構(gòu)酶、PRDX4、CAPG、β-防御素107、微纖絲相關(guān)糖蛋白4等6種蛋白是我們首次采用蛋白質(zhì)組學(xué)技術(shù)成功篩選的腎癌差異表達(dá)蛋白質(zhì),并發(fā)現(xiàn)了1個(gè)預(yù)測(cè)蛋白(UCRIL)。結(jié)論熒光差異雙向凝膠電泳在腎癌組織中獲得較好的篩選效果,新篩選的差異表達(dá)蛋白有望成為腎癌分子生物學(xué)標(biāo)記物,為后續(xù)研究提供了實(shí)驗(yàn)依據(jù)。
[Abstract]:Objective to search for tumor markers associated with renal cell carcinoma (RCC) for clinical diagnosis, drug therapy target selection and prognosis monitoring. Methods fluorescent differential two dimensional gel electrophoresis was used to screen differentially expressed protein spots in 15 cases of renal clear cell carcinoma and adjacent normal renal tissues. T test was used to make quantitative comparison. The ratio of two groups of protein content was 1.5 times and above, P0.05, which was considered to be a differentially expressed protein spot. Differential expression protein spots were identified by matrix assisted laser desorption ionization time of flight mass spectrometry or tandem mass spectrometry. Results A total of 27 differentially expressed protein spots were isolated and 26 proteins were successfully identified by mass spectrometry. 11 proteins were up-regulated in renal cell carcinoma and 15 proteins were down-regulated. Its function covers many aspects of cell activity, including mitochondrial short / branched-chain specific acylCoA dehydrogenase. Aldose 1-epiisomerase, PRDX4,CAPG, 尾 -defensin 107 and microfibril associated glycoprotein 4 are the first differentially expressed proteins in renal cell carcinoma that we successfully screened by proteomics, and a predictive protein (UCRIL). Was found. Conclusion fluorescence differential two-dimensional gel electrophoresis has a good screening effect in renal cell carcinoma tissues. The newly screened differentially expressed proteins are expected to become molecular biological markers of renal cell carcinoma and provide experimental basis for further study.
