根皮素引起前列腺癌LNCaP細胞發(fā)生凋亡的機制研究

【摘要】:根皮素是存在于蘋果、梨等水果及多種蔬菜汁液中的天然活性物質(zhì),具有抗腫瘤活性,能夠有效地誘導細胞凋亡、抑制細胞增殖.本研究初步探討了根皮素誘導前列腺癌LNCaP細胞的凋亡和相關分子機制.通過形態(tài)學觀察、噻唑藍(MTT)比色實驗、CCK-8實驗測定前列腺癌細胞活性;采用DAPI染色法測定細胞核凝縮破裂;運用Annexin V-FITC/PI雙染流式細胞術測定前列腺癌細胞的凋亡率;再通過PI單染流式細胞術測定前列腺癌細胞的周期變化;以及采用Western blot檢測PI3K/AKT信號通路及其下游細胞凋亡相關蛋白和細胞周期相關蛋白的表達.實驗結果表明:根皮素誘導前列腺癌LNCaP細胞周期阻滯在G2/M期,并呈濃度依賴性降低細胞活性、增加細胞核凝縮破裂,顯著提高前列腺癌細胞的凋亡率.在分子機制上,根皮素呈現(xiàn)濃度依賴性下調(diào)PI3K/AKT的活性,從而影響細胞凋亡相關蛋白和細胞周期相關蛋白的表達.因此,根皮素對前列腺癌細胞的生長有顯著的抑制作用,對治療前列腺癌具有潛在的臨床應用價值.
[Abstract]:Phyllostatin is a natural active substance in fruits and vegetables such as apple pear and so on. It has anti-tumor activity and can effectively induce cell apoptosis and inhibit cell proliferation. In this study, the apoptosis of prostate cancer LNCaP cells induced by phylloderin and its related molecular mechanisms were investigated. Morphological observation, (MTT) colorimetric assay, CCK-8 assay, DAPI staining, Annexin V-FITC/PI double staining flow cytometry were used to determine the apoptosis rate of prostate cancer cells. The cell cycle changes of prostate cancer cells were detected by PI single staining flow cytometry, and the expression of apoptosis-related proteins and cell cycle associated proteins in PI3K/AKT signaling pathway and its downstream were detected by Western blot. The results showed that LNCaP cell cycle arrest in G _ 2 / M phase was induced by phylloderin in a concentration-dependent manner, which decreased cell activity, increased nuclear condensation and increased apoptosis rate of prostate cancer cells. In molecular mechanism, hirsutin down-regulated the activity of PI3K/AKT in a dose-dependent manner, thus affecting the expression of apoptosis-related proteins and cell cycle related proteins. Therefore, the growth of prostate cancer cells was significantly inhibited by phylloderin, which had a potential clinical value in the treatment of prostate cancer.
