【摘要】：目的:構(gòu)建體外、體內(nèi)模型,探討羅默堿抗前列腺癌活性。方法:檢測(cè)羅默堿對(duì)多種前列腺癌細(xì)胞(DU145、LNCa P、PC-3和22RV1)增殖、凋亡和遷移的影響,篩選敏感腫瘤細(xì)胞系,構(gòu)建裸鼠荷瘤模型,進(jìn)一步驗(yàn)證羅默堿的體內(nèi)抗腫瘤藥效。結(jié)果:羅默堿對(duì)DU145、LNCaP、PC-3和22RV1細(xì)胞增殖和遷移均具有不同程度的抑制作用,對(duì)其細(xì)胞凋亡亦具有不同程度的誘導(dǎo)作用,其中以LNCaP細(xì)胞最為敏感。裸鼠荷LNCaP腫瘤模型中,羅默堿單獨(dú)干預(yù)組腫瘤重量[(1.99±0.95)g]顯著低于對(duì)照組[(2.95±1.04)g],P0.01;而高劑量羅默堿(30 mg/kg)和紫杉醇聯(lián)合干預(yù)組腫瘤重量[(0.90±0.16)g]均顯著小于其他3組(P0.05或P0.01)。羅默堿單獨(dú)干預(yù)組裸鼠的心臟臟器系數(shù)(0.58±0.06)、肝臟臟器系數(shù)(6.20±0.42)和腎臟臟器系數(shù)(1.49±0.33)均不同程度低于紫杉醇單獨(dú)干預(yù)組(0.66±0.04、6.99±0.72和1.95±0.34,P均0.05),脾臟臟器系數(shù)(0.54±0.11)和胸腺臟器系數(shù)(0.06±0.01)顯著高于紫杉醇單獨(dú)干預(yù)組(0.41±0.09、0.05±0.01,P均0.05)。病理結(jié)果顯示羅默堿干預(yù)組及聯(lián)合給藥組中腫瘤惡變程度和腫瘤轉(zhuǎn)移程度低于對(duì)照組,羅默堿干預(yù)組及聯(lián)合給藥組中小鼠內(nèi)臟損傷明顯小于紫杉醇組水平。結(jié)論:羅默堿發(fā)揮一定的抗前列腺腫瘤功效,同時(shí)聯(lián)合化療藥給藥時(shí)可以降低其毒性。
[Abstract]:Objective: to study the anti-prostate cancer activity of rommerine in vitro and in vivo. Methods: the effects of rommerine on proliferation, apoptosis and migration of various prostate cancer cells (DU145,LNCa PtPC-3 and 22RV1) were detected, and sensitive tumor cell lines were screened to establish tumor-bearing model in nude mice to further verify the anti-tumor effect of rommerine in vivo. Results: rommerine inhibited the proliferation and migration of DU145,LNCaP,PC-3 and 22RV1 cells in different degrees and induced apoptosis in different degrees. LNCaP cells were the most sensitive. In nude mice bearing LNCaP tumor model, the tumor weight of rommerine alone intervention group [(1.99 鹵0.95) g] was significantly lower than that of the control group [(2.95 鹵1.04) g, P 0.01], while the tumor weight of high dose rometin (30 mg/kg) and paclitaxel group [(0.90 鹵0.16) g] was significantly lower than that of the other three groups (P0.05 or P0.01). The heart organ coefficient (0.58 鹵0.06), liver organ coefficient (6.20 鹵0.42) and kidney organ coefficient (1.49 鹵0.33) in rommerine alone intervention group were lower than those in paclitaxel alone intervention group (0.66 鹵0.046.99 鹵0.72 and 1.95 鹵0.34 P 0.05), spleen organ coefficient (0.54 鹵0.11) and thymus organ coefficient (0.06 鹵0.01) were significantly lower than those in paclitaxel alone intervention group (0.66 鹵0.046.99 鹵0.72 and 1.95 鹵0.34 P 0.05). It was higher than that of paclitaxel alone (0.41 鹵0.09 鹵0.05 鹵0.01, P 0.05). The pathological results showed that the degree of tumor malignancy and tumor metastasis in rommerine intervention group and combined administration group was lower than that in control group, and the visceral damage in rommerine intervention group and combined administration group was significantly lower than that in paclitaxel group. Conclusion: rommerine can play a certain role in prostatic tumor and decrease its toxicity when combined with chemotherapeutic agents.
