CircCD44-360與circCD44-594在膀胱癌中的表達(dá)及臨床意義

【摘要】:目的:本研究就circCD44-360(hsa_circ_0021727)與circCD44-594(hsa_circ_0021728)在膀胱癌及癌旁組織中的表達(dá)情況,及它們對CD44基因表達(dá)的影響和相關(guān)性分析進(jìn)行研究,旨在為膀胱癌的診斷和靶向治療提供新的方向。方法:收集膀胱癌及癌旁組織的臨床標(biāo)本,處理標(biāo)本后提取膀胱癌及癌旁組織的總RNA,反轉(zhuǎn)錄獲取目的RNA,進(jìn)行實時定量PCR,檢測circCD44-360與circCD44-594的表達(dá)差異,并分析其分別同親本基因CD44的信使RNA(messenger RNA,mRNA)的關(guān)系進(jìn)而找出其調(diào)節(jié)機制。結(jié)果:1 CircCD44-360與circCD44-594在膀胱癌組織的表達(dá)均上調(diào)。PCR結(jié)果顯示,與癌旁組織相比,circCD44-360與circCD44-594在膀胱癌組織中的表達(dá)水平顯著升高。說明circCD44-360與circCD44-594在膀胱癌組織中的表達(dá)水平均顯著上調(diào)。2 CD44mRNA在膀胱癌組織的表達(dá)上調(diào)。PCR結(jié)果顯示,與癌旁組織相比,CD44mRNA在膀胱癌組織中的表達(dá)水平顯著升高。說明CD44mRNA在膀胱癌組織中的表達(dá)水平均顯著上調(diào)。3 CircCD44-360與circCD44-594均同其親本基因CD44的表達(dá)呈線性正相關(guān)關(guān)系。利用PCR結(jié)果進(jìn)行統(tǒng)計學(xué)分析,circCD44-360與其親本基因CD44的Pearson's correlation線性相關(guān)關(guān)系,R=0.5702,P=0.0087,呈正相關(guān)。另一個環(huán)狀circCD44-594與其親本基因CD44的Pearson's correlation線性相關(guān)關(guān)系,R=0.4474,P=0.0420,呈正相關(guān)。結(jié)論:1 CircCD44-360在膀胱癌組織中表達(dá)上調(diào)。2 CircCD44-594在膀胱癌組織中表達(dá)上調(diào)。3在膀胱癌中,circCD44-360、circCD44-594和CD44的表達(dá)呈正相關(guān)關(guān)系,它們可能通過調(diào)節(jié)CD44基因的表達(dá)促進(jìn)膀胱癌的發(fā)生和發(fā)展。
[Abstract]:Objective: To study the expression of circCD44-360 (hsa_circ_0021727) and circCD44-594 (hsa_circ_0021728) in bladder cancer and para cancer tissues, and their influence on the expression of CD44 gene and the correlation analysis.
