【摘要】：目的初步探討高遷移率族蛋白1(high mobility group protein B1,HMGB1)和腫瘤壞死因子α誘導(dǎo)蛋白3(tumor necrosis factorα-induced protein-3,TNFAIP3)在狼瘡性腎炎(lupus nephritis,LN)患者和人類(lèi)腎小球系膜細(xì)胞(human mesangial cell,HMC)增殖中的作用及機(jī)制。方法收集Ⅳ型LN患者的腎穿樣本,采用免疫熒光和免疫組化技術(shù)檢測(cè)腎小球細(xì)胞中HMGB1、TNFAIP3及IκBα蛋白表達(dá)水平;運(yùn)用人重組HMGB1為刺激物刺激HMC,采用Brd U參入技術(shù)檢測(cè)細(xì)胞增殖水平,并用Western blot技術(shù)檢測(cè)TNFAIP3、IκBα及p65表達(dá)水平。結(jié)果與對(duì)照組相比,LN患者腎小球細(xì)胞中HMGB1及TNFAIP3蛋白表達(dá)升高,IκBα表達(dá)水平降低;體外實(shí)驗(yàn)證實(shí),人重組HMGB1刺激HMC后,細(xì)胞增殖水平明顯升高,同時(shí)在HMGB1刺激30 min后,與對(duì)照組相比,TNFAIP3的蛋白表達(dá)水平明顯升高(P0.05),而IκBα蛋白表達(dá)水平降低(P0.05),隨后p65蛋白表達(dá)明顯升高(P0.05)。結(jié)論 HMGB1和TNFAIP3可能通過(guò)活化NF-κB信號(hào)通路促進(jìn)LN系膜細(xì)胞的過(guò)度增殖。
