自噬減少組蛋白介導(dǎo)的腎小管上皮細(xì)胞凋亡

【摘要】:目的:探討自噬對組蛋白介導(dǎo)的人腎小管上皮細(xì)胞(HK-2)凋亡的效應(yīng)。方法:以不同濃度組蛋白刺激HK-2,應(yīng)用流式細(xì)胞儀和免疫印跡方法檢測HK-2凋亡和自噬相關(guān)蛋白LC3Ⅱ、Beclin1的表達(dá)情況,初步了解組蛋白對HK-2凋亡和自噬的影響。以自噬抑制劑3-甲基腺嘌呤(3-MA)預(yù)處理后,觀察組蛋白誘導(dǎo)的HK-2凋亡和caspase-3活化的情況,進一步明確自噬通路對凋亡的作用。結(jié)果:組蛋白25、50、100μg/mL組HK-2凋亡率和LC3Ⅱ、Beclin1表達(dá)水平均顯著升高(P0.05),并且組蛋白誘導(dǎo)HK-2凋亡具有濃度依賴效應(yīng)。自噬抑制劑3-MA顯著增強HK-2凋亡和caspase-3活化(P0.05)。結(jié)論:自噬能夠減輕組蛋白介導(dǎo)的HK-2凋亡,提示活化自噬通路在急性腎損傷中具有潛在的應(yīng)用價值。
