【摘要】：目的:研究雄性不育相關(guān)基因Boule在小鼠出生后首個精子發(fā)生波中的動態(tài)表達,探討B(tài)oule在精子發(fā)生過程中的調(diào)控作用。方法:運用免疫印跡法分析BOULE蛋白在多組織中的表達情況。運用實時熒光定量、免疫組織化學(xué)和免疫熒光技術(shù)檢測Boule m RNA和BOULE蛋白在雄性小鼠出生后首個精子發(fā)生波各個時間點中的表達。結(jié)果:1免疫印跡法顯示BOULE蛋白在睪丸中特異性高表達,在出生后16 d睪丸可被檢測到,并在隨后的發(fā)育中睪丸和成年睪丸持續(xù)表達;2實時熒光定量方法證實Boule m RNA在雄鼠出生后14 d睪丸中可被檢測到,20 d呈現(xiàn)表達高峰;3免疫組織化學(xué)顯示在精母細胞及圓形精子細胞胞質(zhì)中存在BOULE蛋白陽性信號。結(jié)論:Boule在精子發(fā)生過程中的表達是在發(fā)育水平精密調(diào)控的,在粗線期精母細胞和圓形精子細胞階段的特異表達提示Boule可能在精母細胞進程和圓形精子細胞中發(fā)揮作用。
[Abstract]:Aim: to investigate the dynamic expression of male sterility related gene (Boule) in the first spermatogenesis wave in mice and to explore the regulatory role of Boule in spermatogenesis. Methods: the expression of BOULE protein in multiple tissues was analyzed by Western blot. The expression of Boule m RNA and BOULE proteins in the first spermatogenesis wave of male mice was detected by real-time fluorescence quantitative, immunohistochemical and immunofluorescence techniques. Results the specific expression of BOULE protein was high in testis by Western blotting, and could be detected in testis 16 days after birth. The results of real-time fluorescence quantitative analysis of the continuous expression of Ts2 in the developing testis and adult testis confirmed that Boule m RNA could be detected in the testis of male mice at 14 days postnatal, and the expression peak was detected in the testis of male mice at day 20. Immunohistochemical staining showed that the expression peak was observed in the fine spermatozoa of spermatozoa. Positive signals of BOULE protein were found in cytoplasm of spermatozoa and round spermatozoa. Conclusion the specific expression of Boule during spermatogenesis is precisely regulated at the developmental level. The specific expression of Boule in spermatocytes and round spermatozoa suggests that Boule may play a role in spermatocyte progression and round spermatocyte.
