【摘要】：目的:研究血清脂聯(lián)素水平與前列腺癌發(fā)生的關系以及脂聯(lián)素對前列腺癌細胞生物學特性的影響。方法:采用病例對照試驗回顧性分析45例前列腺癌患者(病例組)和50例非前列腺癌患者(對照組)血清脂聯(lián)素水平和前列腺癌發(fā)生之間的關系。利用慢病毒載體構建脂聯(lián)素受體缺陷的穩(wěn)轉細胞株,用脂聯(lián)素干預前列腺癌細胞系并通過CCK-8實驗、流式細胞實驗、成管實驗和劃痕實驗分析脂聯(lián)素作用升高和降低對PC3細胞生物學行為的影響。結果:與對照組患者[(6.35±4.43)μg/ml]比較,病例組血清脂聯(lián)素水平[(3.49±2.70)μg/ml]降低,并且脂聯(lián)素水平與Gleason評分高低呈負相關(r=-0.504,P0.01)。脂聯(lián)素抑制PC3細胞的增殖,遷移,抑制細胞進入S期,并抑制血管形成,而在脂聯(lián)素受體缺陷的情況下,PC3細胞反而具有更強的增殖、遷移和促血管形成能力。結論:血清脂聯(lián)素下降或脂聯(lián)素受體缺陷可能是促進前列腺癌發(fā)生的因素。
[Abstract]:Aim: to study the relationship between serum adiponectin level and prostate cancer and the effect of adiponectin on the biological characteristics of prostate cancer cells. Methods: serum adiponectin levels in 45 patients with prostate cancer (case group) and 50 patients without prostate cancer (control group) were analyzed retrospectively by case-control trial. A stable cell line with adiponectin receptor deficiency was constructed by using lentivirus vector. Adiponectin was used to interfere with prostate cancer cell line. The effects of adiponectin on the biological behavior of PC3 cells were analyzed by tube forming test and scratch test. Results: compared with the control group [(6.35 鹵4.43) 渭 g/ml], the serum adiponectin level [(3.49 鹵2.70) 渭 g/ml] was decreased, and there was a negative correlation between the adiponectin level and the Gleason score (r = -0.504, P0.01). Adiponectin inhibited the proliferation and migration of PC3 cells, inhibited cell entry into S phase and inhibited angiogenesis. However, when adiponectin receptor was deficient, PC3 cells had stronger ability of proliferation, migration and angiogenesis. Conclusion: the decrease of serum adiponectin or the deficiency of adiponectin receptor may be the factors that promote the development of prostate cancer.
